The incidence of cancer development,invasion and metastasis in cancer biology attentions,especially tumor invasion and metastasis,it is the most critical characteristics of cancer.Urokinase-type plasminigen activator is serine protease.It is through the degradation of the extracellular matrix,promoting tumor invasion and metastasis.It is the abnormal expression of a variety of malignancies,development,invasion and metastasis,can be used as a variety of malignant tumors is an important prognostic indicator.Mainly depend on the growth of tumor blood supply nutrients and oxygen,without blood supply nutrients and oxygen,the tumor will not grow,but the transfer does not occur.The formation of new blood vessels to tumors unlimited,invasive growth provides opportunities and options,making a variety of malignant tumors become more malignant biological behavior.Studies have shown that ovarian cancer has a variety of angiogenic capacity,so increasing the angiogenesis of ovarian cancer by the attention. The abnormal expression of Urokinase-type plasminigen activator is to promote an important factor in tumor angiogenesis.Therefore,the purpose of this study by immunohistochemical methods to detect the expression of uPA and epithelial ovarian cancer angiogenesis,and to investigate patients with epithelial ovarian cancer clinical prognosis.Further explore the expression of uPA in different types of ovarian cancer cells.PURPOSE:1. Explore expression of uPA in epithelial ovarian cancer and distribution.2. Explore the relationship between expression of uPA and microvessel density in epithelial ovarian cancer.Understand the 5-years survival of patients.3. Explore the expression of uPA in different types of ovarian cancer cells.METHOD:1. Immunohistochemical method used to detect uPA in 164 cases of epithelial ovarian cancer and distribution.2. Used immunohistochemistry to investigate the relationship between uPA and MVD.Survey five-year survival of patients3. Western Blotting method used to detect expression of uPA in different types of ovarian cancer cells.4. RT-PCR method used to detect expression of uPA in different types of ovarian cancer cells.RESULT:1. In the four different types of ovarian tissue,expression of uPA in epithelial ovarian cancer is highest. uPA, mainly in the cytoplasm of ovarian tumor cells express some mesenchymal cells, also expressed in vascular endothelial cells, especially in tumor cells and matrix at the junction of uPA was highly expressed.There exists a correlation between expression of uPA and clinicopathological factors of ovarian cancer.2. In epithelial ovarian cancer, new blood vessels around the cancer nest increased significantly. There exists a correlation between expression of uPA and MVD.3. Expression of uPA in ovarian cancer cells with high metastatic cells is more than in low metastatic cells. Expression of uPA in human ovarian cancer with resistant is more than non-resistant.4. Expression of uPA mRNA in ovarian cancer cells with high metastatic cells is more than in low metastatic cells. Expression of uPA mRNA in human ovarian cancer with resistant is more than non-resistant.CONCLUSION:1. In epithelial ovarian cancer,uPA is highly expressed.uPA takes park in ovarian cancer development,invasion and metastasis.2. The high expression promotes epithelial ovarian cancer angiogenesis. uPA and CD34 are very important in epithelial ovarian cancer development,invasion and metastasis. They is estimatede become an important indicator of prognosis.3. There maybe a correlation between uPA and chemotherapy resistance in ovarian cancer.
