Objective:Researched the expresssion level in normal ovarian, benign, borderline and malignant epithelial ovarian tumor tissue of CD73 and MVD.Applied statistics analysis method to study CD73 and MVD expressions in different degree of epithelial ovarian tumor organizations,the relationship with different clinical symptoms,such as histological grade, surgical pathologic staging, lymph node metastasis, ascites and peritoneal flushing fluid,pathological type and patients’ age and other pathological features,as well as the relationship between CD73 and microvessel density. To explore the role of CD73 in the occurrence and development of epithelial ovarian cancer. Maybe CD73 can be a new evaluation index for evaluating the malignant degree and prognosis of epithelial ovarian cancer, and provides a new way for the gene targeted therapy of epithelial ovarian cancer.Methods:Selected the tissue specimens by surgical resection in The China Japan union hospital and Central Hospital of Jilin between January 2014 and December 2015. There are a total of163 cases,including 20 normal ovarian tissues,20 benign epithelial ovarian tumor tissues( which including 10 serous epithelial ovarian tumor tissues and 10 mucinous epithelial ovarian tumor tissues),39 borderline epithelial ovarian tumor tissues(which including 20 serous epithelial ovarian tumor tissues and 19 mucinous epithelial ovarian tumor tissues)and84 epithelial ovarian carcinoma tissues( which including 56 serous epithelial ovarian carcinoma tissues and 28 mucinous epithelial ovarian carcinoma tissues). All specimens were selected for the fresh material, and immediately fixed in 10% formalin solution, after the completion of the Shi Labao buried. The slice thickness was 4 m, staining method: HE staining and immunohistochemical staining of CD73. By immunohistochemistry staining,the positive biopsy kit known as positive control to detect the expression of CD73. At the same time, mark the tumor angiogenesis by CD34 and count the number of MVD.Results:1. Expression of CD73 and MVD in different ovarian tissues,CD73 is widely existed in normal tissues, and is mostly expressed in mesenchymal cells.CD73 in normal ovarian tissues are all low expression, but in benign, borderline and malignant epithelial ovarian tumor tissues are high expression, whose rates increased gradually.statistical analysis can find: comparison between borderline group and benign group, P > 0.05, with statistical difference; malignant group and benign group were compared, with significant difference(P< 0.001). MVD count was also gradually increased, and the four groups were compared with each other, all with statistical difference, P<0.001.2. The CD73 expression level and MVD count in epithelial ovarian carcinoma have some relationships with clinical pathological features. The aged less than 50 years and >50years were compared with each other, statistically significant differences between the expression level of MVD and CD73 count was not significant(P>0.05); the pathologic stage I ~ II and III ~ IV CD73 expression level in patients was compared with each other, P<0.01,with significant statistical difference. The count of MVD comparison, P<0.001, has a statistically significant difference; the histological grade of G1+G2 group and G3 group compared the expression of CD73 and MVD count, P<0.01, with statistical significance;according to the lymph node metastasis as grouping criteria to compare CD73 expression and MVD count, P<0.001, was statistically significant; as ascites and cancer cells in peritoneal lavage fluid or not, the expression of CD73 and MVD count was compared,P<0.01, there were statistical significance; the comparison of pathological types are divided into viscous and serious group, between the two groups, the expression of CD73 and MVD parity count, P>0.05, no significant difference.3.In patients with epithelial ovarian cancer, the higher degree of expression of MVD,the greater of CD73. By Spearman rank correlation analysis, there was a positive correlation between the expression of both(r=0.914, P< 0.001), and the expression level of CD73 was correlated with MVD.Conclusion:1. The positive expression of CD73 and MVD in epithelial ovarian cancer was significantly higher than that in normal ovarian, benign or borderline epithelial ovarian tumors. It suggested that the upregulation of CD73 expression and MVD may be closely related to the occurrence of ovarian epithelial cancer.2. The positive expression of CD73 and MVD are positively related to various pathological parameters, which indicates that the high expression of CD73 is related to the malignant degree, such as clinical pathological stage, histological differentiation degree,lymph node metastasis, ascites and peritoneal washing fluid with the tumor cells.They have closely relationship with the development of the disease and the prognosis of epithelial ovarian cancer.3. The high expression of CD73 has positive correlation with MVD in ovarian cancer,which indicates that the role of CD73 in the metastasis of epithelial ovarian cancer may be accomplished by promoting tumor angiogenesis.In summary, CD73 in epithelial ovarian cancer is highly expressed, indicating that it plays a role in the pathogenesis of tumor; CD73 expression and degree of malignancy degree could be used as indicators of new assessment of disease progression and prognosis of ovarian epithelial cancer and new ideas of gene targeted therapy.
