Objective:To study the expression of Ki-67 and Bcl-2 protein in oral squamous cell carcinoma and evaluate their significance in pathogenisis and development of oral squamous cell carcinoma(OSCC).Clinical data and methods:The expression of Ki-67 and Bcl-2 protein were detected by PV-6000 immunohistochemical staining method in paraffin embedded slices of 50 oral squamous cell carcinomas, in which,35 cases were collected with the adjacent OSCC mucosa and the normal tissues together, and all the patients did not receive any anticancer therapy,and analyzed their clinicopathologic significance with respect to histological differentiation, tumor size, lymph node metastasis and clinical stage.Results:The positive expression rate of Ki-67 was 98%(49/50) in OSCC, but it was weak positive or negative in normal mucosa. The positive expression of Ki-67 was correlated with histological differentiation, tumor size and clinical stage (P<0.001,P<0.05,P<0.05),but was not correlated with lymph node metastasis (P>0.05). The positive expression rate of Bcl-2 was 94%(47/50) in OSCC, the positive correlation between Bcl-2 expression and the differentiation was statistically significance (P<0.001), but no correlation was formed to the lymph node metastasis, tumor size and clinical stages.Conclusion:The expression of Ki-67 and Bcl-2 might be used as important biological markers for oral squamous cell carcinoma, and both Ki-67 and Bcl-2 were the major gene proteins for promotion of cancer cell proliferation and apoptosis inhibition.
