The desired of human tumor animal model can provide a scientificplatform for the development of clinical anticancer drugs and newtreatment method. In recent years, human-derived orthotopic tumor modelhas become the most extensive and most valuable application of moderncancer research model, the simulation of the human body tumor can be agood natural growth process and biological behavior. This model is the useof human tissue, or tumor cells directly transplanted in nude miceformation of the corresponding organs and tumors. Objective and Significance To Use tumor cell lines and human tumortissues established in nude mice by Orthotopic transplantation with human. In the modeling process, several experiments nude mice back of the neckskin and varying degrees of hyperkeratosis of the phenomenon. Analysis ofabnormal causes for this anomaly in the model, as well as analysis of theimpact of the phenomenon of tumor model. Stable and reliable model forthe development of anticancer drugs.Research methodsThe first part, the establishment of Orthotopic transplantation of ahuman tumor model in nude mice1. Using human breast cancer MDA-MB-231-Luc cell suspension nudemouse mammary fat pad in situ injection, the establishment of the cellsuspension in situ COI (Cellular Orthotropic Injection of Cell Suspensions)model. Logarithmic phase of human breast cancer MDA-MB-231-Luccells collect in vitro, the number of effective live cells>95%, cut theright side of the chest second on the breast skin of exposed fat pad, and slowly injected into the cytosol after suture wound. Mice in vivo imagingdevice to monitor the in situ tumor and the production of tumor growthcurves and mice body weight change curve modeling the success of theabove indicators to measure.2. Selection the surgical removal of tumors in the liver cancer patients, removing the necrotictissue and Congestion, cut into appropriate size of thetumor after treatment, the establishment of the intact tissue orthotopicimplantation of SOI (Surgical Orthotropic Implantation of HistologicallyIntact Tumor Tissue) model. Anesthesia the animal, then abdominalpuncture needle for direct inoculation of tumor tissue in the liver. To stanchbleeding and suturing the wound after tumor inoculation. When thetumor-bearing mice up to1000mm3, tumor-bearing rats were euthanized byCO2, remove the tumor piece passage between rodents, and production ofoffspringin nudemice, tumor growth curves and bodyweight changecurve, measuring the quality of the model. The second part, the analysis of the causes for hyperkeratosis in nudemice after Tumor transplantationIn breast cancer tumor model established by the first14days,17days ofthe liver tumor model,5and12animal models hyperkeratosis phenomenon, skin white dander.Normal animals were quarantined and observed clinical symptoms, abnormal skin tissue of nude mice model Gram staining microscopicexamination of bacteria culture, culture detection of the pathogenicinfection in nude mice, Histopathological examination. Environmentmonitoring and fall air monitoring of bacteria and micro-organisms.Statistical incidence of animal, analysis of tumor growth and weightchanges rate for abnormal animal tumor models and normal animal tumormodels, compare the significant. The results and conclusionsSuccessful use of surgical methods of establishment of Orthotopictransplanted human tumors in nude mice of COI and SOI model stability subcutaneous tumor passage. Established a solid tumor models, andprovide basic parameters for tumor growth.About3-7days nude mouse body weight gradually returned tonormal, the phenomenon of hyperkeratosis was disappearance, the tumorcontinued to grow. Cultured Gram-positive strains isolated in nude miceand no strong pathogenicity; histological examination showed nosignificant pathological changes. Nude mouse does not occur againhyperkeratosis. The statistical analysis of the abnormal tumor animalmodels and normal tumor animal models of tumor size and weight changedifference, the difference was not significant. Description of the symptomsof the transition horny no significant effect on the establishment of thenude mouse tumor model. Hyperkeratosis caused by modeling animalphenomenon may be due to individual differences of the animals, or carrysome kind of condition unknown pathogens.
