Objective To observe the relationship between expression of TNF-a,IL-10 and Brain Edema at the surrounding of hemorrharic of intracerebral hemorrhage in rats and the Intervention Role of injection of Milkvetch Root. approach the effect of injection of Milkvetch Root on Brain Edema and the expression of TNF-a,IL-10 after intracerebral hemorrhage and provide the experimental and theory support about injection of Milkvetch Root on the patient of intracerebral hemorrhage. Methods The ICH model in rats was established by heparinizedâ…¦-type collagenase into caudate nucleus of rats, and then observe the effects of injection of Milkvetch Root on it. 180 rats were randomly divided into three group: sham operated group,ICH group and Milkvetch Root group. Every group was divided into 6 h,24 h,48,72 h,7 d five time points. Every time points have twelve rats. Berderson score to evaluate behavioral change, the dry/wet weight method to check up cerebral water content, pathological change at the surrounding of hemorrharic of intracerebral hemorrhage was observed under lightmicroscop of HE staining, the expression of TNF-a,IL-10 at the surrounding of hemorrharic of intracerebral hemorrhage were detected by immunohistochemistry. Results The effect of injection of Milkvetch Root on the score of behavioral change after ICH in rats: the neurologic impairment appeared in the ICH group and the Milkvetch Root treated group after the beginning of the ICH,and gradually aggravated. the score was highest at 48h,and then gradually recovered,It has recovered the normal at 7d .The score in the Milkvetch Root treated group was lower than it in the ICH group at 48h and 72h(P<0.05), the difference between this has statistical significance; The effect of injection of Milkvetch Root on the cerebral water content after ICH in rats: the cerebral water content begin to increase in the ICH group and the Milkvetch Root treated group after the beginning of the ICH, to reach peak at 48h, and then gradually decreased, to recover the normal at 7d. the cerebral water content in the Milkvetch Root treated group was lower than it in the ICH group at 48h and 72h(P<0.05), the difference between this has statistical significance; The effect of injection of Milkvetch Root on pathological change after ICH in rats: Tissue at the surrounding of hemorrharic of intracerebral hemorrhage have obviously inflammatory reaction after ICH in rats, small amounts inflammatory cell begin to infiltrate at 6 h after ICH, the infiltrating of neutrophil inflammatory cell gradually increased at 24 h, to reach peak at 48h ,peripheral glia cell begin to increased at 72h, hematoma obviously deflate, inflammatory cell gradually disappeard, peripheral glia become more obvious at 7 d. Extent of brain tissue edema and infiltrating of inflammatory cell in Milkvetch Root group were more soften than ICH group at same time point,thats more clearly at 48h;The effect of injection of Milkvetch Root on TNF-a after ICH in rats: A low level of TNF-a expression were detected at the surrounding of hemorrharic in the ICH group and the Milkvetch Root treated group at 6h. and gradually increased, the expression reach highest at 24h,then gradually decreased .the expression in the Milkvetch Root treated group was lower than it in the ICH group at 6h,24h,48h,72h,7d. the difference between this has statistical significance; The effect of injection of Milkvetch Root on IL-10 after ICH in rat: A low level of IL-10 expression were detected at the surrounding of hemorrharic in the ICH group and the Milkvetch Root treated group at 6h. and gradually increased, the expression reach highest at 7d. the expression in the Milkvetch Root treated group was lower than it in the ICH group at 6h,24h,48h,72h,7d. the difference between this has statistical significance.Conclusion The score of behavioral change after ICH coincide with the the cerebral water content .prompted Brain Edema is the main reason which resulted in the neurologic impairment. The cerebral water content and the infiltrating of inflammatory cell coincide with the expression of TNF-a. prompted TNF-a can promote Inflammatory reaction,aggravate Brain Edema. It is opposited with the expression of IL-10.prompted IL-10 can restrain Inflammatory reaction,lessen Brain Edema. The injection of Milkvetch Root can decrease the expression of TNF-a and increased the expression of IL-10.it also can decrease the infiltrating of inflammatory cell,restrain Inflammatory reaction lessen Brain Edema,improve the neurologic impairment.it can provide effective help for treating the cerebral injury.
