Objective: To investigate the expression of MMP-2, VEGF, CD105 and Ki67 in human small cell lung cancer tissues, normal lung tissues and the correlation of the four indicators, and analyze the relationship between the biological characteristics and prognosis. It will provide theoretical basis for the clinical treatment and prognosis in SCLC.Methods: Immunohistochemistry SP method was used to detect the expression of MMP-2,VEGF,CD105 and Ki67 in 42 cases of SCLC paraffin histopathological tissues and 8 cases normal lung tissues. SPSS17.0 statistical software was used for statistical analysis, and p-value less than 0.05 were considered significant. The correlation between the four indicators and the major clinical characteristics of SCLC were analyzed by chi-square test. Postoperative survival period analysis were computed by the Kaplan-Meier method and compared by the Log-rank test. The prognostic relevance of the four indicators and conventional prognostic factors were analyzed by COX regression model.Result:1. The positive expression rate of MMP-2, VEGF, CD105 and Ki67 in 42 cases of SCLC tissues was 50.0%, 66.7%, 81.0% and 64.3%, respectively. Positive expression rate of MMP-2, VEGF, CD105 and Ki67 in normal lung tissues was 12.5%, 12.5%, 0% and 0%, respectively. The different positive rates between SCLC and normal lung tissues had statistical significance (P<0.05). 2. The expression of MMP-2,VEGF,CD105 and Ki67 was not correlated with gender, age and smoke statu(sP>0.05). 3. The expression of MMP-2,VEGF and CD105 was correlated with tumor size, lymph node metastasis , distant metastasis and clinical stage(P<0.05 or P< 0.01 ) .Ki67 expression had no significant correlation with the features above-mentioned(P>0.05) but lymph node metastasi(sP<0.05). 4. The expression of MMP-2,VEGF was correlated with CD105 and Ki67(P<0.05 or P<0.01).5. There were significant positive correlations between MMP-2 and VEGF in SCLC(P<0.05,r=0.447). 6. The positive expression rate of MMP-2, VEGF and CD105 in PFS≥6 month group was higher than the PFS<6 month group(P<0.05 or P<0.01). 7. The expression of MMP-2, VEGF, CD105 and Ki67 was was correlated with the prognosis of SCLC. In additional, tumor size, lymph node metastasis, distant metastasis, clinical stage were another prognosis factors. 8. The COX regression demonstrated the expression of MMP-2, VEGF, tumor size, lymph node metastasis, distant metastasis and clinical stage were independent prognostic indicators of SCLC.Conclusion:1. The positive expression rate of MMP-2, VEGF, CD105 and Ki67 are significantly higher in SCLC than the normal lung tissues. This result showed that MMP-2, VEGF, CD105 and Ki67 may play an important role in the occurrence and development of SCLC, and also could be used for diagnosis.2. MMP-2, VEGF and CD105 expression was correlated with tumor size, lymph node metastasis, distant metastasis and clinical stage.Ki67 was only correlated with lymph node metastasis. This result suggested that the four indicators were closely correlated to the invasion and metastasis of SCLC .3. The expression of MMP-2, VEGF was correlated with CD105, which showed that both the two indicator could participate the angiogenesis in SCLC.4. The expression of MMP-2, VEGF was correlated with Ki67.It suggested that MMP-2, VEGF may promote the proliferation of SCLC, simultaneously.5. There were significant positive correlations between MMP-2 and VEGF in SCLC, and it showed that MMP-2 and VEGF can promote the angiogenesis in SCLC by synergistic effect.6. The correlation between MMP-2, VEGF, CD105 expression and TTP demonstrate that MMP-2, VEGF and CD105 could be the indicators of recurrence in SCLC.7. The expression of MMP-2, VEGF, CD105 and Ki67, tumor size, lymph node metastasis, distant metastasis and clinical stage were correlated to the prognosis in SCLC, but only MMP-2, VEGF expression, tumor size, lymph node metastasis, distant metastasis and clinical stage were the independent prognostic indicators.
