Objective:Make the migraine model by subcutaneous injection of nitroglycerin. To observe the influence of TouTong Ning on contents ofβ-EP in blood plasma and expressions ofβ-EP, c-fos,c-jun,5-HT in brain tissue of the rats with migraine. To investigate the possible mechanism of TouTong Ning in prevention and cure migraine.Methods: 1. 72 SD rats weighted 200~250g were devided into 6 groups randomly: NS control group, model group, flunarizine group,Tou Tong Ning high dose group,TouTong Ning medium dose group,TouTong Ning low dose group.There are 12 rats of each group. Each group after modeling was randomly divided into 4-hour and 6-hour groups .There are 6 rats of each group.2.Make the migraine model animal by subcutaneous injection of nitroglycerin.3.Medication: NS control group and model group were intragastric administration by physiological saline (0.5ml/100g) everyday each for six days. Flunarizine group were intragastric administration by flunarizine suspensions (1.04mg/kg ) everyday each for six days. Tou Tong Ning groups were intragastric administration by Tou Tong Ning suspensions respectively(0.76/kg,0.38/kg,0.19/kg) everyday each for six days. In the first seven days: model group,flunarizine group,Tou Tong Ning groups were injected subcutaneouly nitroglycerin by 10 mg/kg to make the migraine model. NS group was injected subcutaneouly physiological saline by 2ml/kg. 30 minutes after injection each group were intragastric administration respectively one time.4.To observe the behavioral symptoms in each group.5.To detect the contents ofβ-EP in the blood plasma by radioimmunuoassy.6.To detect expressions ofβ-EP,c-fos,c-jun,5-HT in brain tissue with immunohistochemical method.Results:1.The results of behavioral symptoms in each group: The model group appeared redness both ears,scratching head with forelimb frequently,climbing cage,dysphoria phenomenons.The phenomenons continued with l-3 hours. The NS group only in a few hours scratched head and climbed cage a few times sporadic, and becamed normal after. They didn't appear redness both ears and other phenomena of irritability.The Tou Tong Ning groups and flunarizine group appeared redness both ears,scratching head also,but terminal times of redness both ears and scratching head were earlier than the model group,which was statistical significance compared with the model group(P<0.05).Scratching head and climbing the cage were obviously decreased compared with the model group,which were statistical significance compared with the model group(P<0.05).2.The results ofβ-EP contents in blood plasma in each group: The contents ofβ-EP in blood plasma in the model group in two-time points have decreased significantly. The contents of 6-hour group was lower than the 4-hour group. There were statistical significance compared with the NS group (P <0.05).The contents ofβ-EP in two- time points had obviously increased in Tou Tong Ning groups and flunarizine group. There were statistical significance compared with the model group(P<0.05).3.The results ofβ-EP protein's expressions in the hypothalamic of rats in each group: Theβ-EP positive cells slightly increased in the hypothalamic of rats in two-time points in model group. The positive cells of 4-hour group were more than the 6-hour group. There were statistical significance compared with the NS group (P <0.05).The expressions ofβ-EP positive cells in the hypothalamus in two- time points had obviously increased in Tou Tong Ning groups and flunarizine group.There were statistical significance compared with the model group(P<0.05). 4.The results of c-fos, c-jun protein's expressions in the periaqueductal gray of rats in each group: The c-fos, c-jun positive cells increased significantly in the periaqueductal gray of rats in two-time points in model group. The expressions of 4-hour group were more than the 6-hour group. There were statistical significance compared with the NS group (P<0.05).The expressions of c-fos,c-jun in the periaqueductal gray in two- time points had obviously decreased in Tou Tong Ning groups and flunarizine group. There were statistical significance compared with the model group(P<0.05).5.The results of c-fos, c-jun protein's expressions in the hypothalamic of rats in each group: The expressions of c-fos,c-jun positive cells increased significantly in the hypothalamic of rats in two-time points in model group. The expressions of 4-hour group were more than the 6-hour group. There were statistical significance compared with the NS group (P <0.05).The expressions of c-fos,c-jun positive cells in the hypothalamus in two- time points had obviously decreased in Tou Tong Ning groups and flunarizine group. There were statistical significance compared with the model group(P<0.05).6.The results of 5-HT protein's expressions in the midbrain of rats in each group: The expressions of 5-HT positive increased slightly in the midbrain of rats in two-time points in model group. The expressions of 4-hour group were more than the 6-hour group. There were statistical significance compared with the NS group (P <0.05).The expressions of 5-HT positive cells in the midbrain in two-time points had obviously increased in Tou Tong Ning groups and flunarizine group. There were statistical significance compared with the model group(P<0.05).Conclusion:1.Tou Tong Ning can improve the behavioral symptoms of the rats with migraine by nitroglycerin. Among them, Tou Tong Ning high dose group and TouTong Ning medium dose group.are more obvious role than the TouTong Ning low dose group. 2.Tou Tong Ning can restrain the protein's expressions of c-fos and c-jun in the periaqueductal gray and the hypothalamus of the rats with migraine by nitroglycerin. Among them, Tou Tong Ning high dose group and Tou Tong Ning medium dose group.are more obvious role than the TouTong Ning low dose group.3.Tou Tong Ning can up regulation the contents ofβ-EP in blood plasma and increase the protein's expressions ofβ-EP in the hypothalamus and 5-HT in the midbrain of the rats with migraine by nitroglycerin.Among them, Tou Tong Ning high dose group and TouTong Ning medium dose group.are more obvious role than the TouTong Ning low dose group.
