ObjectiveTo study the positive expression of Transgelin in CRC,meanwhile detect the relationships of the expressions of Transgelin,MMP-9,CEA and their clinical and pathological data. Then to explore the role of the combined expressions of Transgelin,MMP-9,CEA take in the predictation and prognosis of CRC.So as to offer theory basis to the finding of new CRC marks.MethodsThe expression of Transgelin,MMP-9,CEA are detected by immunoh- istochemistry S-P method in 56 colon carcinoma (experimetal group) and 10 normal tissue besides the colon carcinoma tissue (control group).These 56 patients with CRC underwent surgical treatment from 2000.1 to 2009.10 at the 88th hospital of PLA and their clinical and pathological data are kept complete.We use the S-P to analyze the positive rate of the expressions of Transgelin,MMP-9,CEA in CRC. Using SPSS13.0 to do the statistical analysis,including the simple expression of single protein and Logistic multifactor to analysis ,including the simple expression of single protein and Logistic multifactor to analyze whether the expression of Transgelin is the monofactor of CRC.Result1.The trangelin is expressed positively in cytoplasm of normal colorectal tissue, the positive rate is 90%(9/10 examples). It expressed strongly in well-differentiated CRC, the positive rate is 85.7%(24/28 examples). It expressed moderately in differentiated CRC, the rate is 55%(11/20 examples). And it expressed slightly or cannot be expressed in low-differentiated CRC, the rate is only 12.5%(1/8 examples). There is a significance in statistically when comparing among these groups.(P﹤0.01)2.The MMP-9 is expressed positively in cytoplasm of normal colorectal tissue, the positive rate is 10%(1/10 examples). It expressed strongly in well-differentiated CRC, the positive rate is 60.7%(17/28 examples). It expressed moderately in differentiated CRC, the rate is 70.0%(14/20 examples). And it expressed slightly or cannot be expressed in low-differentiated CRC, the rate is only 87.5 %(7/8examples). There is a significance in statistically when comparing among these groups.(P﹤0.01)3.The CEA is expressed positively in membrane or cytoplasm of normal colorectal tissue, the positive rate is 0%(0/10 examples). It expressed strongly in well-differentiated CRC, the positive rate is 46.4%(13/28 examples). It expressed moderately in differentiated CRC, the rate is 55%(11/20 examples). And it expressed slightly or cannot be expressed in low-differentiated CRC, the rate is only 37.5%(3/8 examples). There is a significance in statistically when comparing among these groups.(P﹤0.01)4.Analysis the relationship of Transgelin,MMP-9 and CEA under single factor level, using the multivariate logistic method to analysis whether the Transgelin protein is an independent risk in CRC invasion or lymph node metastasis.Conclusion:1.Transgelin protein were low to undetectable levels in colorectal carcinoma. The express of transgelin in peri-tumor tissue showed strong positive,To provide more information for clinical practice.2. The express of Transgelin,MMP-9,CEA protein was no correlation with age,sex,location of tumor,tumorous diameter .3.There is a correlation in the express of Transgelin,MMP-9 proteins in colorectal carcinoma.to depth of invasion and metastasis.They can be used as parameters for invasion and metastasis.4. There was an inverse correlation between Transgelin and MMP-9 proteins expression in colorectal carcinoma. The expressions of transgelin proteins are independent risk factors to the invasion or lymphatic metastasis. in colorectal carcinoma.
