Objective:To observe the anti-inflammatory injury therapy effect and the content of IL-6, IL-10 and the expression of T-lymphocyte subsets in rats with experimental ulcerative colitis with the herb Kuijiequan Retention-enema. To discuss its therapeutic mechanism.Methods:40 male SD rats were randomly divided into the normal group, the model group, the SASP conrol group and the Kuijiequan group, containing 10 rats in each group. Ulcerative colitis model was established in SD rats by injecting the mixing of 2,4,6-trinitro-benzene sulfonic acid(TNBS) and 50% alcohol into anus, except for the normal group. the 3th day after model establishment, start drug intervention for 10 continuous days. Diarrhea, bloody stool and the change of weight was observed. The macroscopic and histological chan-ges of the colon were evaluated. the content of IL-6 and IL-10 in the serum of rats was detected by enzyme-linked immuno sorbent assay(ELISA). the expression of T-lymphocyte subsets in peripheral blood of rats was determined by flow cytometry(FCM).Results:The model group of rats appeared severe diarrhea and bloody stool, the weight loss of which was obvious; Colon mucous membrane in the model group had serious erosion, ulcer, damaging of glandular organ, high inflammation grade. Compared with that in normal group, the content of IL-6 in the serum of model group of rats was significantly increased and the content of IL-10 was obviously decreased; There was no obvious change in percentage content of peripheral blood CD3+CD4+ cell between the normal group and the model group; The percentage content of peripheral blood CD3+CD8+cell in the model group was lower than that of the normal group and the CD3+CD4+/CD3+CD8+ ratio was significantly increased. Com-pared with the model group, the symptoms of the SASP control group and the Kuijiequan group had improved. The colon tissues have different levels of inflammation repairing and ulcer healing. the serum IL-6 was decreased, and IL-10 was increased obviously:but had no obvious change in CD3+CD4+cell, the CD3+CD8+cell was rised and the ratio of CD3+CD4+/ CD3+ CD8+was decreased; There has no significant difference of other index between the SASP control group and the Kuijiequan group.Conclusion:The herb Kuijiequan Retention-enema can effectively treat the rats with experimental ulcerative colitis, improves the general condition and alleviate the inflammatory damage of colon. It has the effects of reducing the content of IL-6 and increasing IL-10, and also can increase the percentage content of peripheral blood CD3+CD8+cell, reducing the radio of CD3+CD4+/CD3+CD8.Thus we deduce that its probably plays a role of immunoregulatory action by regulating the balance of pro-inflammatory/anti-inflammatory cytokine and modify the proportion of T-lymphocyte subsets.
