| Objective:Ulcerative Colitis is a kind of chronic non-specific inflammatorydisease which has an unknown etiology and occurred in the colon, the distalcolon, and also throughout the entire colon to erosion, ulcer as the mainpathological changes toabdominal pain, diarrhea, mucus pus and blood, andtenesmus as the main clinical manifestations. In recent years, with the changeof people's living habits and environmental factors, the incidence of UCincreased year by year, seriously affecting patients with learning, working andliving. During the long Clinical Practice we selected an experiencedprescription named Xiezhuojiedu Decoction based on Turbidity Toxin theoryof traditional chinese medicine to treat this disease with the method of purgingturbidity and ascending lucidity,detoxification and removing stasis.throughanimal experiments, observation Turbidity detoxification of UlcerativeColitisrat intestinal mucosal EGF, NO, NOS and serum the EGF, NO and NOSexpression, and sulfasalazine effects to explore the possible mechanism ofaction by the party's treatment of UC, UC treatment in order to provide acertain amount of theory and practicebasis.Methods:2,4,6-trinitrobenzene-sulfonic acid,TNBS/ethanol combinativemethod of establishing model used to Induce animal models in Rat UlcerativeColitis in this study,45healthy and clean wistar Rats were taken,which weremale, weighting220--250g.8rats were randomly chosen to be the controlgroup, the other rats were given30%Ethanol Solution which contains30mgTNBS, to make Ulcerative Colitis animal models. Three days later,5rats wererandomly chosen to be killed to take their colon models. If there are hype-remias, edemas and ulcers, the UC rats model making can be thought to besuccessful. The other rats were randomly divided into4groups with8rats ineach group, and they are separately named modeling group,SASP group, Xiezhuojiedu Decoction high dose group,Xiezhuojiedu Decoction low dosegroup. The control group,modeling group rats were given normal saline bygavage.The rats of SASP group,Xiezhuojiedu Decoction low dose group,Xiezhuojiedu Decoction high dose group were separately given SASP at0.3g/kg, water decoction of Xiezhuojiedu Decoction at10.0g/kg and20.0g/kgby gavage,Once a day for14days and observed the effect. I studied anddiscussed the experiment from three aspects: In Experiment1, to observed theeffect of Xiezhuojiedu Decoction on general condition and pathological effecton Ulcerative Colitis Model Rats; In Experiment2, to observed the effect ofXiezhuojiedu Decoction on the expression of EGF of the colonic mucosa onUlcerative Colitis Model Rats; In Experiment3,to observed the effect ofXiezhuojiedu Decoction on NO and NOS acivity using the nitrate reductasemethod and chemical method. the data were analysised with SPSS13.0.Two-tailed test with P﹤0.05were considered significant.Results:1Experiment1showed:Compared with the control group, the rats' general condition changeddifferently in modeling group and all the treatment groups. All those mentalstatus, activity condition, hair glossiness, appetite, weight defecatecharacter,ect. were better in Xiezhuojiedu Decoction high dose group andSASP group than the other groups,model group was the worst.The rats'disease active index (DAI):BG is the lowest in DAI score in allgroups.MG's DAI score significantly higher compared with the BG. The datahad statistical significance between two groups (P<0.05). comp-ared with theMG,DAI score of HG,LG and SG were significantly decre-ased,Data hadstatistical significance(P<0.05). compared with the HG, DAI score of SGdecreased, Data had statistical significance(P<0.05). compared with the LG,DAI score of SG data had no statistical significance(P>0.05).The rats'colonic mucosa damage index (CMDI):BG is the lowest in CMDIin all groups.MG's CMDI significantly higher compared with the BG. Thedata had statistical significance between two groups (P<0.05). compared with the MG, CMDI of HG,LG and SG were significantly decreased,Data hadstatistical significance(P<0.05). compared with the HG, CMDI of SGdecreased, Data had statistical significance(P<0.05). compared with the LG,CMDI of SG data had no statistical significance(P>0.05).histomorphology observation of rats' colon under the light microscop-e: The BG rat colon structure were clear,there were no erosions andhyperemia,mucosal epithelial cells arranged in neat rows,the intestinal glandin right rules and goblet cells were rich, there were capillaries texure in thelamina propria,The SG and LG group colon ulcers were more superficial ulcernext to the epithelial repair significantly active gland hyperplasia, crypt,intestinal glangs and stroma can be seen a small amount of inflammatory cellinfiltration,The HG group colonic mucosa epithelium and glandular arrangedin neat rows,active glands,goblet cells,interstitial congestion and edema is notobvious,crypt,intestinal glands and interstitial inflammatory cells infiltration,its basic form was sim-ilar to the BG group.2Experiment2showed:to observe the effect of Xiezhuojiedu Decoction on EGF on UlcerativeColitis Model Rats:Compared with the BG, MG group was no differentces inEGF(P>0.05). Compared with the MG, The EGF in SG group was significa-ntly higher,The data had statistical signifycance between two groups (P<0.05).compared with the LG, The EGF in SG group was no differences,Data had nostatistical significance(P>0.05). compared with the SG, The EGF in HGgroup was significantly higher,The data had statistical signifycance betweentwo groups (P<0.05).2Experiment3showed:to observe the effect of Xiezhuojiedu Decoction on serum NO and NOSactivity on Ulcerative Colitis Model Rats: Compared with the BG, serum NOand NOS activity in MG group was significantly higher,The data had statistic-al signifycance between two groups (P<0.05). Compared with the MG, serumNO and NOS activity in SG group was reduced,The data had statistical signif-ycance between two groups (P<0.05). Compared with the SG, LG group was no differentces in serum NO and NOS activity(P>0.05). Compared with theSG, serum NO and NOS activity in HG group was reduced,The data hadstatistical signifycance between two groups (P<0.05).to observe the effect of Xiezhuojiedu Decoction on colon tissue NO andNOS activity on Ulcerative Colitis Model Rats: Compared with the BG, colontissue NO and NOS activity in MG group was significantly higher,The datahad statistical signifycance between two groups (P<0.05). Compared with theMG, colon tissue NO and NOS activity in SG,LG and HG were reduced,compared with the MG, colon tissue NO and NOS activity in HG group wassignificantly reduced,The data had statistical signifycance between two groups(P<0.05). Compared with the MG, colon tissue NO and NOS activity inLG,SG was no differentces(P>0.05).Conclusion: Xiezhuojiedu Decoction can improve the rats' general conditi-on and reduce the pathological damage score,promote the expression of EGFin the serum and colon tissue,while reducing NO and NOS expression in theserum and colon tissue. the protective effect of EGF on colonic mucosal barri-er and promote the repair of mucosal injury, accelerate the healing of UC col-onic mucosa lesions and adjust the colonic mucosa of immune function.thismay be one of the mechanisms Xiezhuojiedu Decoction treatment of UC.Ourstudy the clinical treatment of UC may be help us found a new opportunity. |
