Objective:Ulcerative Colitis is a kind of chronic non-specific inflammatory diseasewhich occurred in the colon with a long course, easy to repeated attacks, andhas a certain relationship with the incidence of colon cancer. In recent years,the incidence of UC increased year by year, seriously affecting patients withlearning, working and living. Xiezhuojiedu recipe is an effective experienceprescription we have selected during the long Clinical Practice,which is basedon Turbidity Toxin theory of traditional chinese medicine to treat this diseaseand pathogenesis of ulcerative colitis. Through animal experiments in thisstudy, we want to explore the possible mechanism of its treatment of UCthrough observing the influence of this method on ulcerative colitis serumIL-4, IL-6, and comparing with sulfasalazine, in order to explain themechanism for the treatment of ulcerative colitis with modern medical theoryMethods:In this study, there were45weight220-250g, male, healthy and cleanWistar rats,8of them were randomly selected as the blank group (BK), theremaining rats copy the animal models of ulcerative colitis withTrinitrobenzene sulfonic acid (2,4,6-trinitrobenzene-sulfonic acid, TNBS)/ethanol joint modeling method. Three days later, five model rats were killed,to check for the colon specimens and confirm that there was congestion,edema, ulcer formation and pathological changes. After the modeling, theremaining32rats were randomly divided into: the model group (MK),sulfasalazine group (SK), Xiezhuojiedu low-dose group (LK), Xiezhuojieduhigh-dose group (HK), n=8. The BK group normally feeded,MK groupgavage with saline, SK, LK, HK group were given sulfasalazine liquid0.3g/kg,Xiezhuojiedu recipe10.0g/kg Xiezhuojiedu recipe20.0g/kg of daily irrigation stomach,the treatment lasting for14consecutive days to evaluateefficacy. The experimental study investigated two aspects:1to observe theimpact of Xiezhuojiedu recipe on the general condition and colonichistopathology of the rats with UC;2to observe the influence of this methodon ulcerative colitis serum IL-4, IL-6. SPSS13.0was used for statisticalanalysis with P <0.05as statistically significant criteriaResults:1The general condition and colonic histopathology of the rats with UCGeneral condition: compared with BK group, the general condition ofMK group and the treatment group rats were different; the mental state,reaction capability, hair gloss, food intake, body weight, stool situation of HKbetter than the other groups;SK and LK group similar to each other;the situation of MK group was the worstRat disease activity index (DAI) score: compared with BK group rats, theDAI score in MK, SK, LK,HK group increased, the difference wasstatistically significant (P<0.05);the DAI score in SK, LK, HK group rats werelower, the difference was statistically significant (P<0.05) as compared withthe MK.;compared with SK, the DAI score in HK rats decreased significantly(P<0.05); the DAI score in LK rats had no significant difference with SK (P>0.05);compared HK with LK,the DAI score in HK was lower than the LKgroup (P <0.05).Colon mucosa damage index (CMDI) Rating: compared with BK grouprats, the CMDI ratings in MK, SK, LK HK rats increased, the difference wasstatistically significant (P<0.05);the CMDI score in SK, LK, HK group ratswere lower, the difference was statistically significant (P<0.05) as comparedwith the MK.;Compared with SK, the CMDI score in HK rats decreasedsignificantly (P<0.05);the CMDI score in LK rats had no significant differencewith SK (P>0.05); Compared HK with LK,the CMDI score in HK was lowerthan the LK group (P <0.05).Histomorphology observation of rats’ colon under the light microscop-e:BK rats colon tissue structure was clear, with no congestion、erosion and ulceration, mucosal epithelial cells arranged in neat rows, Midgut glandsstructured; the MK rats mucous layer missing, showed the varying degrees ofcongestion, edema, erosion, ulceration; in the of SK and LK, colon ulcersshallow, a small amount of inflammatory cell infiltrationed crypt intestinalglands and interstitial; the stromal of HK had no significant congestion andedema, scattered inflammatory cell infiltrationed the crypts, intestinal glandsand interstitial2The serum IL-4, IL-6levels of the rats with UCSerum IL-4levels: compared with the BK group, the IL-4levels in serumof the other groups were lower, the difference was statistically significant (P<0.05); compared with MK group, the IL-4levels in serum of SK、 HK、andLK group were elevated, the difference was statistically significant(P <0.05).comparing between the LK group and SK group, the difference was notstatistically significant (P>0.05); the IL-4levels in serum of HK increasedcompared with the SK group, the difference was statistically significant (P<0.05); compared to LK, the IL-4levels in serum of HK group increased, thedifference was statistically significant (P <0.05).Serum IL-6levels: compared with the BK group, the IL-6levels in serumof MK, SK, LK and HK were elevated, the difference was statisticallysignificant (P<0.05); compared with MK, the IL-6levels in serum of SK,LKand HK group were lower, the difference was statistically significant (P<0.05). comparing between LK and SK group the difference of the IL-6levelsin serum was not statistically significant (P>0.05); compared with the SKGroup, the IL-6levels in serum of HK group decreased, the difference wasstatistically significant (P<0.05), compared with LK, the IL-6levels in serumin HK group increased, the difference was significant difference (P<0.05).Conclusion:Xiezhuojiedu recipe could improve the general condition of the rats withulcerative colitis and reduce the pathological damage score, increase the IL-4levels in serum, reduce the IL-6levels in serum. This may be one of themechanisms why Xiezhuojiedu recipe could treat UC. It found a new opportunity for our future clinical research on UC.
