Objective: Ulcerative colitis(Ulcerative Colitis is called UC) or chronic non-specificity ulcer coliti is an unknown inflammatory bowel disease UC is recommended to be one of the refractory diseases by World Health Organization.which have the characteristics of longer courses andrecurrence,and has a certain relationship with colon cancer' attack. Xiezhuojiedu Recipe is a remarkable effective therapy of my teacher,which has recived good curative effects. Through the research and experiment, this study is to observe Xie zhuo jie du Recipe's effects on UC and demonstrate its immune functional mechanism.Methods: Forty healthy SD rats were chosen and randomly divided into five groups: blank control group(BK),model group(MK), Sulfasalazine group(LK),Chinese medicine high dose group (GK),Chinese medicine low dose group (DK).The rats in BK were fed nomally,the rats in other 4 groups were given 30﹪ethanol enema liquid by Enema which was mixed with TNBS(50mg/kg-1) to make UC animal models. The rats in BK were fed normally,the rats in MK were given normal saline by garage, the rats in LK were given SASP by gavage ,the rats in GK and DK were separately given Xiezhuojiedu Recipe with high dose and low dose. Then we observed the effects. The experiment contained three components. Experiment I was about Xiezhuo jiedu Recipe's effect on T cell subset and the complement. Experiment II was about Xiezhuojiedu Recipe's effect on the immune globulin .Experiment III was about Xiezhuojiedu Recipe's effect on the expression of ICAM-1 .Results:Experiment I:1.1 The general condition (DAI)Compared with MK(2.016±0.401), GK(0.429±0.153), DK (0.496±0.144), LK(1.286±0.299)and BK(0.378±0.133)all could significantly decrease the General state in UC rats (P<0.01); Compared among DK,GK,LK,DK was significantly lower than LK(P<0.01).1.2 Colon tissue Pathological damageThe rats'colonic mucous layer was deleted in large area in LK, with infiltrating of inflammatory cells in submucosa, hyperemia, edema, ulcer focus and granulation tissues;the rats'mucosal epithelium and crypt arranged regularly in GK, the basic form was similar to normal tissues; the rats'hyperemia, edema were not significant in DK,LK,but we could see a few inflammatory cells intersperse.1.3The T cell subset(%)CD4+:Compared with MK(46.56±4.72), GK (24.04±4.28), DK(38.38±4.89), LK(40.75±6.67) and BK (24.76±4.89) all could significantly decrease the percentage of CD4+ (P<0.01);And compared CD4+ content in blood among GK,DK and LK, GK was significantly lower than LK(P<0.01),DK and LK had no significant defferences(P>0.05);GK was significant than DK(P<0.01).CD8+:Compared with MK (17.21±1.57), GK(24.04±4.28), BK(30.48±4.38) and LK(22.89±4.56) all could significantly increase the percentage of CD8+(P<0.01);DK(21.22±6.27)did not have significant variation(P>0.05); compared among DK,GK,LK, DK and LK have significant defferences, GK and DK did not have significant defferences;Compared with DK,GK is significantly higher.Experimen II:2.1 The immune globulinIgA: Compared with MK (0.136±0.097), GK (0.012±0.015), BK (0.022±0.018) and LK (0.063±0.047) all could significantly decrease the content of IgA (P<0.01);Compared among GK,DK and LK,DK was significantly lower than LK(P<0.05); DK and LK did not have significant defferences (P>0.05);compared with DK, GK could significantly decreased the content of IgA(PDK-LK<0.01).IgG: Compared with MG (0.169±0.082), DK (0.204±0.068) , GK (0.210±0.085) and BK (0.169±0.082) all could significantly decrease the content of IgG (P<0.01);compared among GK,DK and LK, GK was significantly lower than LK(P<0.05);DK and LK did not have significant defferences (P>0.05);compared with DK, GK could significantly decrease the content of IgG (P<0.05).IgM: Compared with MK(0.606±0.183), BK(0.380±0.112) Could significantly decrease the content of IgM (P< 0.05);Compared among the others: GK(0.589±0.210), DK (0.524±0.126), LK(0.579±0.178)did not have significant the variation( P>0.05).2.2 The complement C3,C4C3: Compared with MK (0.106±0.013), BK (0.186±0.013),GK (0.310±0.131), DK(0.216±0.036) and LK(0.229±0.046)all could significantly increase the content of C3 (P<0.01, P<0.05);and compared among DK,GK and LK, GK was significantly higher than LK (P<0.05) , DK didn't have significant defferences;compared with DK, GK could significantly increase the content of C3 (P<0.05) .C4:Compare with MK (0.016±0.005),GK (0.106±0.028) ,DK (0.043±0.016) and LK (0.071±0.038) all could significantly increase the content of C4 (P<0.01,P<0.05);and compared among GK,DK and LK,GK was significantly higher than LK(P<0.05) , DK and LK did not have significant differences(P>0.05);compared with DK, GK could significantly increase the content of C4 (P<0.05).Experiment III:The expression of the Intercelluar adhesion molecule-1Compared with MG(5.750±1.282), GK(2.954±0.676),LK (3.560±0.473),BK(3.031±0.334) all could significantly decrease the expression of the Intercelluar adhesion molecule-1 (P<0.01);DK could significantly decrease the expression of the Intercelluar adhesion molecule-1 (P<0.05);compared among GK,DK and LK, GK was significantly better than LK(P<0.05);GK and DKdid not have significant defferences(P>0.05).Conclusions:1 Xiezhuojiedu Recipe has good preponderance in treating UC and can improve UC rat's general condition and colonic mucosa pathology,especially GK's integrity effect is significantly better than LK.2 Xiezhuojiedu Recipe's effect maybe:it can increase the content of C3andC4, lessen the immune response of IgG,IgA in blood and decrease the expression of the Intercelluar adhesion molecule-1. This Recipe can also adjust the balance between CD4+ and CD8+.
