Objective: Ulcerative colitis is a kind of chronic non-specificinflammatory diseases mainly related to intestine, colonic mucous membrane,the digestive system diseases of difficult cured which has complex etiologyand longer course. During a large number of clinieal praetice, we selected anexperienced preseription named Xiezhuojiedu Decoction based on TurbidityToxin theory of traditional chinese medicine,it has reliable clinical effect. Thepurpose of this study was, through the animal experiment, to observe effects ofXiezhuojiedu decoction on ulcerative colitisrat intestinal mucosal theCyclooxygenases-2and ProstaglandinE2expression, clarify the possiblemechanism of action by the decoction, in order to further explore thepathogenesis and clinical treatment into more meaningful to provide theoryand practice basis.Methods: In the study,40healthy and clean wistar Rats were taken,weighing220~250g and were randomly divided into five groups with8rats ineach group: the blank group (BG), the model group (MG), the sulfasalazinegroup (SG), Xiezhuojiedu Decoction high dose group (HG), XiezhuojieduDecoction low dose group (LG).2,4,6-trinitrobenzene-sulfonic acid,TNBS/ethanol combinative method of establishing model used to induceanimal models in rat ulcerative colitis.3days after modeling, the blank groupand model group fed with normal saline by gavage, and the other group ratswere treated for14days. We observed the changes of mental status, bodyweight, general changes in stool, etc.; evaluation of colonic mucosa damagedegree; using immunohistochemical staining of rat colon mucosa COX-2,PGE2expression. Validity evaluation based on the observed results, analyzingXiezhuojiedu Decoction on COX-2and PGE2effects. Results:1The general stateCompared with the BG, the rats’ general condition changed differently inMG and other the treatment groups. All those mental status, activity condition,appetite, hair glossiness,ect. in Xiezhuojiedu Decoction high dose group, werebetter than other treatment groups.2The general observation on the rats.colon and colonic mucosa damageindex(CMDI)By the visual observation of rat’s colon mucosa, there were no congestion,edema, colonic wall thickening, ect in BG. We could observe colon mucosaedema, bleeding, erosion, ulcer, colon bowel wall thickening and expansion inMG. After treatment, the above situation of the LG, HG and SG all alleviated.3The observed under optical microscopeCompared with the BG, each group of rats’ mucosa condition changeddifferently in epithelial integrity degree, gland structure, inflammatory cells,ect. MG colon mucosa layer is missing, and large number of inflammatory cellin submucosa, we could visible acute vasculitis pathological in bottom of theulcer lesions.After drug intervention, the treatment group rats colon mucosapathological changes all have varying degrees of ease, which HG groupvisible improved mucosal gland hyperplasia, lined duct,interstitial scatteredsmall amount of inflammatory cells, abundant goblet cells, the degree ofrecovery is close to the BG.4The test results of COX-2and PGE2Compared with the BG(0.229士0.002), the expression of colon tissueCOX-2in MG(0.5070.243)increased significantly, had significant difference(P<0.05); Compared with the MG, the expression of colon tissue COX-2inHG(0.228士0.017), SG(0.311士0.007) and LG(0.315士0.006) decreasedobviously, had significant difference(P <0.05); Compared with the LG, SG, theexpression of COX-2in HG decreased obviously, had significant difference(P<0.05); Compared with the SG, the expression of COX-2in LG didn’t havesignificant difference(P>0.05). Compared with the BG(0.115士0.002), the expression of colon tissuePGE2in MG (0.452士0.006) increased significantly, had significantdifference(P <0.05); Compared with the MG, the expression of colon tissuePGE2in HG(0.118士004), SG(0.202士0.007) and LG(0.203士0.004)decreased obviously, had significant difference(P <0.05); Compared with theLG, SG, the expression of PGE2in HG decreased obviously, had significantdifference(P <0.05); Compared with the SG, the expression of PGE2in LGdidn’t have significant difference(P>0.05).Conclusions:Xiezhuojiedu Decoction can significantly improve thegeneral condition of UC rats, and decrease colonic mucosa damage index. Theexperiment proved Xiezhuojiedu Decoction had good curing effect on theUC rats. Xiezhuojiedu Decoction can reduce the COX-2and PGE2expressionlevel, consequently inhibit inflammatory cells,repair the damaged intestinalmucosa, accelerate the ulcer healing, regulating immune function. Thetherapeutic mechanism is probably related to inhibition of COX-2,down-regulated the PGE2expression level.
