Dynamic Study Of Role Of Highly Active Antiretroviral Therapy On Peripheral Level Of Interleukin-7, CD127and Interleukin-16in Patients With HIV Infection And AIDS

Objective:To explore the dynamic changes and it's significance of peripheral blood level of CD4+T lymphocyte, IL-7, CD127and IL-16during HAART treatment in patients with HIV infection and AIDS. To provide scientific theoretical basis for illustrating in-depth immune pathogenesis and establishing therapy strategy for HIV infection and AIDS patients.Methods:Twenty HIV/AIDS patients who received HAART therapy were tested for CD4, CD8lymphocytes by Flow Cytometry and the leveles of IL-7, CD127, IL-16in peripheral blood by ELISA. At four time points (before treatment, after treatment12weeks,24weeks and48weeks), at the same time, fifteen healthy controles were tested the same itemes for comparison. SPSS17.0packages were used in the analysis of the data.Results:(1) The counts of CD4+T lymphocytes at24weeks after treatment were significantly increased (P<0.001), comparison the12weeks after treatment and the before treatment. The counts of CD4+T cells at after treatment24weeks,12weeks and before treatment were significantly lower than health controls (P<0.001).(2) The level of IL-7in peripheral blood at24weeks after treatment were significantly lower (P<0.05), comparison the12weeks after treatment and the before treatment, the level of IL-7in peripheral blood at after treatment24weeks,12weeks and before treatment were significantly increase than health controls (P<0.05).(3) The level of CD127in peripheral blood at24weeks after treatment were significantly lower (P<0.05), comparison the12weeks after treatment and the before treatment, the level of CD127in peripheral blood at after treatment24weeks,12weeks and before treatment were significantly increase than health controls (P<0.05).(4) The level of IL-16in peripheral blood at24weeks after treatment were significantly increased (P<0.05), comparison the12weeks after treatment and the before treatment, the level of IL-16in peripheral blood at after treatment24weeks,12weeks and before treatment were significantly increase than health controls (P<0.05).(5) The level of IL-7and CD4+T, SCD127and CD4+T, IL-16and CD4+T cells were no correlation (P>0.05) at before treatment. The level of IL-7and SCD127have moderate correlation (P<0.05, r=0.624), at before treatment. The level of IL-7and IL-16have moderate correlation (P<0.05, r=0.631).(6) The level of IL-16in patient's peripheral blood after treatment is influenced by ages.Conclusion:(1) In HAART early, the patient's immune system had a certain extent restore.(2) During HAART process, the level of IL-7and CD127decreases gradually, with also can't be recovered to normal, suggesting IL-7system may play an important role in HIV/AIDS patient's immune reconstitution.(3) During HAART process, the level of IL-16increases gradually, suggesting IL-16in HIV/AIDS patient's condition observed and as a factor of adjuvant therapy has broad application prospects.(4) There is a positive relationship between IL-7and IL-16, It provide theory basis for IL-7and IL-16joint adjuvant therapy with HIV/AIDS.
