Objective: Malignant tumor is a serious impact on human health andlife.The number of patients die each year from cancer around the world is anupward trend. How to better understand and control the occurrence anddevelopment of cancer is a difficult problem of the medical profession. Gastriccarcinoma is a common malignant tumor in China. About17million peopledied of gastric carcinoma each year in China, almost close to1/4of all cancerdeaths, and each year there are20000new patients with gastric carcinomaappears, in our country, the incidence rate in the various types of tumors is thetop. Gastric carcinoma is a serious threat to people's life. But the exactpathogenesis of gastric carcinoma is not clear. With the wide application ofmolecular biology techniques, the occurrence and development of gastriccarcinoma research from the molecular genetics level is gradually becomingthe focus of the medical profession.TIP30gene is a newly discovered tumor metastasis suppressor gene,TIP30gene are expressed in a variety of tissues in the human, but decreased orabsent in a variety of tumor tissue. Tip30is found in the study of vitrotranscription of the human immunodeficiency virus (HIV). Sequence analysisshowed that Tip30and tumor metastasis suppressor gene CC3is the sameprotein. In1997, using RNA differential display technology found that CC3isa metastasis suppressor gene.KAI-1gene is a tumor metastasis suppressor gene, it can regulate theinteraction between the cell and the extracellular matrix of cell, also betweenthe cells and cells,the tumor cells away from the primary tumor becomes verydifficult. Normal expression of the KAI-1protein play an inhibitory effect inmany tumor metastasis process. Various studies have shown that the KAI-1gene's deletion, mutation or abnormal expression played an important role in the malignant tumor invasion and metastasis.In this experiment by detecting the expression of TIP30and KAI-1ingastric carcinoma, adjacent tissues and normal gastric mucosa tissue,wediscussed the relationship between the TIP30and KAI-1,and discussed therole of TIP30and KAI-1in the development of gastric carcinoma and therelationship between clinic pathological characteristics,provide guidance andassistance for the early diagnosis and clinical treatment of gastric carcinoma.Methods: Immunohistochemical(streptavidin-peroxidase, SP) techniquewas applied to detected the expression of TIP30and KAI-1in45cases ofgastric carcinoma,45cases of the proximal adjacent tissues and25cases ofnormal gastric mucosa tissue, and the comprehensive analysis of combinedwith clinic pathological factors including the patient's age, gender, infiltrationdegree, lymph node metastasis and so on. Using the statistical softwareSPSS13.0statistical data processing, count with χ~2test, P <0.05hasstatistically significant. It analyzed the relationship between the expressionboth in gastric carcinoma by using Spearman rank correlation which is one ofnon-parameter statistics.Results:1The expression of TIP30and KAI-1in gastric carcinoma tissues, adjacenttissues and normal gastric mucosa tissue1.1There are22cases which TIP30protein expression was positive in the25cases of normal gastric mucosa, the positive rate was88%(22/25); there are29cases which TIP30protein expression was positive in the45cases of adjacenttissues, the positive rate was64%(29/45); there are18cases which TIP30protein expression was positive in the45cases of gastric carcinoma tissues,the positive rate was40%(18/45). In normal gastric mucosa tissue,the positiveexpression rate of TIP30was significantly higher than the adjacent tissues andthe difference was significant (P <0.05),the positive expression in the adjacenttissue was higher than the gastric carcinoma tissues,the difference wassignificant(P <0.05).1.2There are20cases which KAI-1protein expression was positive in the25 cases of normal gastric mucosa, the positive rate was80%(20/25); there are24cases which KAI-1protein expression was positive in the45cases of adjacenttissues, the positive rate was53%(24/45); there are14cases which KAI-1protein expression was positive in the45cases of gastric carcinoma tissues,the positive rate was31%(14/45). In normal gastric mucosa tissue,the positiveexpression rate of KAI-1was significantly higher than the adjacent tissues andthe difference was significant (P <0.05),the positive expression in the adjacenttissue was higher than the gastric carcinoma tissues,the difference wassignificant(P <0.05).2The relationship between the expression and the clinic pathologic features ofTIP30and KAI-1both in gastric carcinoma tissues2.1The positive expression of TIP30in differentiated and well-differentiatedgastric carcinoma tissues was68.75%(11/16),the positive expression rate inpoorly differentiated gastric carcinoma tissues was24.14%(7/29), thedifference was significant(P <0.05). The positive expression rate of TIP30ininfiltrating serosa of the gastric carcinoma tissues was23.81%(5/21), thepositive expression rate in not invading the serosa was54.17%(13/24),thedifference was significant(P <0.05). The positive expression rate of TIP30ingastric carcinoma tissues with lymph node metastasis was16.67%(4/24), thepositive expression rate of TIP30in gastric carcinoma tissues without lymphnode metastasis was66.67%(14/21),the difference was significant(P <0.05).The positive expression of TIP30had no significant relationship with patient'sage and sex.(P>0.05).2.2The positive expression of KAI-1in differentiated and well-differentiatedgastric carcinoma tissues was62.5%(10/16),the positive expression rate inpoorly differentiated gastric carcinoma tissues was13.79%(4/29), thedifference was significant(P <0.05). The positive expression rate of KAI-1ininfiltrating serosa of the gastric carcinoma tissues was14.29%(3/21), thepositive expression rate in not invading the serosa was45.83%(11/24),thedifference was significant(P <0.05). The positive expression rate of KAI-1ingastric carcinoma tissues with lymph node metastasis was12.5%(3/24), the positive expression rate of KAI-1in gastric carcinoma tissues without lymphnode metastasis was52.38%(11/21),the difference was significant(P <0.05).The positive expression of KAI-1had no significant relationship with patient'sage and sex.(P>0.05).3The relationship of expression between TIP30and KAI-1in gastriccarcinomaUsing Spearman rank correlation analysis indicated that the TIP30andKAI-1expression in gastric carcinoma was positively correlated (rs=0.627, P<0.05)Conclusions:1The expression of TIP30in normal gastric mucosa tissue showed highexpression and it was closely related to the degree of tumor differentiation,depth of invasion and lymph node metastasis. The expression of TIP30indifferentiated and well-differentiated gastric carcinoma tissues was higher thanthat in poorly differentiated gastric carcinoma tissues;The expression ofTIP30in without serosal invasion in gastric carcinoma tissues wassignificantly higher than that in serosal invasion;The expression of TIP30ingastric carcinoma tissues with lymph nodes metastasis was significantly lowerthan in gastric carcinoma tissues without lymph nodes metastasis.2The expression of KAI-1in normal gastric mucosa tissue showed highexpression and it was closely related to the degree of tumor differentiation,depth of invasion and lymph node metastasis. The expression of KAI-1indifferentiated and well-differentiated gastric carcinoma tissues was higher thanthat in poorly differentiated gastric carcinoma tissues;The expression ofKAI-1in without serosal invasion in gastric carcinoma tissues wassignificantly higher than that in serosal invasion;The expression of KAI-1ingastric carcinoma tissues with lymph nodes metastasis was significantly lowerthan in gastric carcinoma tissues without lymph nodes metastasis.3The expression of TIP30and KAI-1in gastric carcinoma tissues waspositively correlated and both make an important role in the developmentof gastric carcinoma. TIP30and KAI-1may be closely related to the development, invasion, metastasis and prognosis of gastric carcinoma, thejoint evaluation of their function has important clinical significance.
