In this study, the chemical constituents of 75% EtOH extracts from northeast Astragalus mongholicus Bunge were studied. 10 compounds were isolated, the structures of 9 compounds were identified as astragaloside IV and calycosin. The determination method of astragaloside IV and calycosin in Radix Astragali with HPLC and the HPLC fingerprints of Radix Astragali were developed for the quality assessment of Radix Astragali. The assaying method was simple and accurate; with good reproducibility for the quality assessment for Radix Astragali.PCR direct sequencing was applied to direct the ITS region sequences and matK gene sequences of Northeast Radix Astragali. The phylogenetic tree of ITS region sequences and matK gene sequences from Radix Astragali reconstructed by neighbor-joining method shew the phylogenetic relationship of Radix Astragali was conformity.The systematics of Northeast Radix Astragali was studied in botanic, chemistry and molecular biology. With the results of 3 species of northeast Radix Astragali in morphous and distribution area, cluster analysis of HPLC fingerprints of the raw materials, phylogenetic tree of ITS and matK sequences. We got the conclusion that the Astragalus mongholicus Bunge should be a species as Astragalus membranaceus (Fisch.) Bunge, and the Astragalus mongholicus Bunge var. borealimongolicus (Y. Z. Zhao) Q. Sh. Sun et J. H. Tu comb. nov. should be a variation of Astragalus mongholicus Bunge.
