How to evaluate the quality of traditional Chinese medicine effectively has always been the tremendous problem troubling the traditional Chinese medicine research areas.That is because Chinese herbs,using in guide of traditional Chinese medicine theory,are very different from chemical medicines with clear structures and specific efficacy. Even a single Chinese herb always has many kinds of functions and components,which leads the complexity of the quality evaluation. So it's not scientific enough to evaluate the Chinese herbs'qualities with only one kind of components.However, it's hard to find a perfect way to evaluate them all-around at present. In fact, it may be more reasonable to evaluate the quality of the medicinal herbs with a specific function. Accordingly, we make Rhubarb as our investigational object, which has a plenty of functions and components, and make a research on the relationship among species, producing areas, curtivars and its components through HPLC, matK sequences, and cDNA-SRAP.The research mainly acquired following achivements:1The work succeeded in establishing three methods for four sorts of characteristic component contents of Rhubarb using HPLC.2Through the content results of samples from different species, producing areas, curtivars of Rhubarb,we find that:(a) Rheum tanguticum Maxim,ex Balf. cultivators from RuoErGai have the highest content of Catechin, about1.5-5times higher than the other samples,(b) Wild Rheum laciniatum samples from XiaHe and a wild Rheum officinate sample from HuaShan have the highest content of gallic acid,about5?12times higher than others,(c) The content of Sennoside B is about5times higher than the content of Sennoside A in wild Rheum laciniatum samples,while the other have about2times higher content of Sennoside A than the content of Sennoside B.(d) Rheum palmatum samples from QuanShui have a low content of Sennoside, especially the R-QS5sample which even can't detect by HPLC.(e) In general, wild samples have a higher content Sennoside than the cultival ones,(f) Wild Rheum laciniatum samples from XiaHe and a wild Rheum officinate sample from HuaShan are special according to the contents of the four sorts of characteristic components, which can draw the similar conclusion from matK sequence analysis,(g) Rheum palmatum samples from QuanShui all have similar contents of all the components, but bad quality, which suggests that their origins may be the same,(h) Rheum palmatum samples from KangDing are similar to Rheum tanguticum samples, which may mean the ones are potential good samples. 3From matK sequence analysis,we find that different species, producing area, and cultivars can be divided by SNPs of matK sequence.4From the same area samples'component contents, we find that there's nearly no difference between4cultural year samples and5cultural year samples except Sennoside. And every component content in radix is more than that in rhizoma. Meanwhile, we select0610?0701,0613?0713,0605?gs2samples as the following experiment objects.5Through cDNA-SRAP,we got9unknown protein gene pieces, which may help to discover some new functional genes.All in all,through our work, we have found the different sorts of rhubarb according to their functional components, which may give a good example to evaluate the quality of Chinese herbs.What's more, we have discovered9unknown protein gene pieces, which can give a basis to select molecular marker and functional genes.
