Rhizoma Chuanxiong which is a umbellate family plant is Ligusticum chuanxiong Hort.’s dried root and stem. It has a long medicinal history in China and its effects are very significant. It can active blood and relieve pain and in the clinical treatment. It is mainly used for cardiovascular diseases and gynecological diseases. Many Traditional Chinese Medicine contain Rhizoma Chuanxiong.Volatile oil is an important active ingredient of Rhizoma Chuanxiong.The study was carried out with L9(34) orthogonal design, using three indexes including the Dujiangyan Rhizoma Chuanxiong particle size, soaking time.the added water as the investigation factors, to evaluate the volatile oil extraction rate, then choose the best volatile oil extraction technology. Namely, particle size was20-50mesh.added8times water, soaking time was4hour, extraction time was5hour. Obtaining enough volatile oil through the best volatile oil extraction technology.And then do research on the purification of volatile oil.Volatile oil is an important active ingredient of Rhizoma Chuanxiong.but Rhizoma Chuanxiong volatile oil volatilizes easily and is sensitive to light and temperature and is susceptible to oxidize,which affects the stability of its preparation.In this study,the method of saturated water solution was used to prepare β-CD inclusion complex of volatile oil. The study was carried out with L9(4) orthogonal design, using three indexes including the proportion of volatile oil to β-cyclodextrin, inclusion temperature and stirring time as the investigation factors.to comprehensively evaluate the volatile oil utilization efficiency.inclusion yield and inclusion oil content,then choose the best preparation technology and validate the inclusion compound.Study results show that the optimum preparation conditions for inclusion were established as: volatile oil of Rhizoma Chuanxiong:β-CD was1:8(mL:g),the temperature was at45℃and the stirring time was2h.After validation,inclusion has been packaged successfully,and established method was stable and reliable and has a higher rate of inclusion.It was suitable for improvement of preparation which contains Rhizoma Chuanxiong. In recent years, the demand for Rhizoma Chuanxiong is increasing in the international market and foreigners pay attention to its heavy metals gradually. So establishing the heavy metals of Rhizoma Chuanxiong determination methods and analyzing it are very necessary. At present, determination methods of heavy metals contain ICP-AES、ICP-MS、AAS、AFS and so on.The content of Cu in the8different Rhizoma Chuanxiong samples solution was determined by FAAS, Cd and Pb were determined by GFAAS. Values of RSDs (n=6) found for Cu, Cd and Pb were all less than2.5%. Test for recovery was made by standard addition method, giving values of recovery in the range of100.0%-108.3%. The established method met the requirement.The results refer to"the medicinal plants and preparation of import and export of green industry standard",which has the limit of heavy metals. In addition to the content of Cu of the eighth sample was exceeded, other content of heavy metals of Rhizoma Chuanxiong met the requirement. The method was rapid, accurate and suitable for the detection of bulk samples, this study also provided some reference for Rhizoma Chuanxiong quality control in actual production.
