Diosgenin was an important basic material for the production of hormone dr?gs.Dioscorea zingiberensis C.H.Wright containded the most diosgen in the world.Diascin desugared to obtain diosgenin.The traditional production process of diosgenin used a mass of sulfuric acid which will making Serious environmental pollution.The purpose of this article was to research a cleaner production process of diosgenin.Domestic researching on enzymatic conversion of diosgenin was less,Especially the snailase.This experiment used ultrasonic cell crushing technology to acton the Dioscorea zingiberensis C.H.Wright soaked by ethanol,and then acided hydrolysis diosgenin.Comparing with cellulase and Snailase.worked together to make diosgenin.Optimization of ultrasonic crushing process and enzyme process,Aiming at puting forward a new technology of making diosgenin by ?ltrasonic-assisted compound enzyme The main research contents and results were as follows:1?The technology of Using ultrasonic assisted acid hydrolysis extraction: On the basis of single factor experiment,optimize ing the ultrasonic crushing process,The influence factors that affect the yield of diosgenin from large to small was in turn: ultrasonic power > ultrasonic temperature > ultrasonic time.The best process was extraction power of 570 W,extracting time of 45 min,extracting temperature of 70 ?.The yield of validation experiment with diosgenin was 0.8802%,higher than that of ethanol hot reflux extraction by 19.28%,than direct acidolysis extraction was increased by 30%.2?Cellulse and snailase were used to prepare dios genin of Dioscorea zingiberensis C.H.Wright.After determining the key steps in the operation and single factor experiment,then optimiz the digestion process.The influence factored that affect the yield of diosgenin from large to small was in turn: enzymatic hydrolysis temperature >enzyme dosage> enzymatic hydrolysis p H > enzymolysis time.The optimal condition for enzymatic hydrolysis were as follows: enzymolysis time of 48 h,enzyme dosage of 8%,enzymatic hydrolysis of pH 6.0,hydrolysis temperature of 50oC.Thus,the yield of diosgenin prepared under those condition was up to 0.5249%.3??ltrasonic auxiliary compound with enzymatic hydrolysis,increased 25% than the direct enzymatic hydrolysis,the yield was the same with the traditional acid hydrolysis.The entire didn't need to consume alcohol and acid.It's a kind of high efficiency low energy,economy and environmental protection of the preparation method of diosgenin.4?The optimum extraction conditions was determined after the hydrolysis :Using 90-120? of petroleum ether extracts 6 hours in 90 ? water bath pot.5?A novel HPLC method was established to determine diosgenin of Dioscorea zingiberensis C.H.Wright.Diosgenin was separated by WondaSilC18(5? m,4.6*150 mm)column with acetonitrile-water(80:20)as mobile phase,and the UV detector was set at 203 nm.The method in 4.70 ?g/mL to 940 ?g/mL keeps good range linear,Precision,stability,repeatability and recovery for the experimental relative standard deviation is within the errorrange.
