Pseudosciaena crocea is a kind of pseudosciaena,which belongs to Queenfishes. It is one of four fishery economic fishes in our country.Ninde Fujian is chief producing area of Pseudosciaena crocea,which the annual output of Pseudosciaena crocea has close to100thousand tons.Egg of Pseudosciaena crocea was always discarded as waste and can’t be fully used. This paper use egg of Pseudosciaena crocea which was from ninde fujian as raw material,lipid composition and composition of fatty acids were extractde and analysis,extraction technology of Phospholipid were studie,the compositions of Phospholipid of fish egg was analyzed and determinded.On the basis of the fundamental research on it,the anti-oxidantive effect of Phospholipid from the egg of Pseudosciaena crocea was studied.The main contents in this paper were concluded as follow:1.The lipid composition and composition of fatty acids of the egg of Pseudosciaena crocea were analysised,the results showed that the fat content of wet basis and dry basis were19.6%and44%,the proportions of phospholipids and cholesterol were61.2%and3.2%. Moreover,the egg of Pseudosciaena crocea contain abundant co-3PUFA unsaturated fatty acid.The total content of EPA and DHA in lipid reach to25.5%.The determination results can provide theoretical basis for exploitation of ω-3PUFA Phospholipid from the egg of Pseudosciaena crocea.2. An extraetion process of Phospholipid from the egg of Pseudosciaena crocea was studied.The egg of Pseudosciaena crocea was homogenated,draw off neutral lipid and pigment by acetone,after that,ethanol was used as the solvent to extract Phospholipid.Different factors were considered.Andorthogonal tests were done based on the single factor tests.It was indicated that the optional conditions for Solvent Method was:solid-liquid ratio of acetone and fish egg was1:9,extracting40min,after pumping filtration,the filter cake was extracted by92%ethanol,the solid-liquid ratio1:8,extracting temperature45℃,extracting20min and the frequency was3times. Under these conditions,the extraction rate could reach to47.73%.Ultrasonic on the extraction rate of Phospholipid was studied,the results showed with ultrasonic power 28KHz,the extraction rate could reach to59.37and was increase24.39%comparing with simple solvent extraction.3.The method of Phospholipid extraction was protease enzymolysis applied to extract Phospholipid from the egg of Pseudosciaena crocea.Influences of enzymatic hydrolysis was researched by single factor experiment.On the basis of single factor experiment,In order to study the relationships between the extraction rate of Phospholipid(Y) and hydrolysis temperautre(X1), hydrolysis time(X2) added enzyme qunatity(X3), the orthogonal rotation combination design was employed.The results showed that the optional conditions for Solvent Method was:using papain,solid-liquid ratio was1:2,pH value6.0, hydrolysis temperautre45℃,hydrolysis time1.5h,added enzyme qunatity125U/g (raw material).After hydrolysis,centrifuging,precipitation extracted by ethanol(combined with the optimum process of Solvent Method) Under these condition,the extraction rate could reach to70.92%and was increase48.59%comparing with the solvent extraction.4.Using TLC combined with spectrophotometry to test the components of Phospholipid.The results showed that the Phospholipid from the egg of Pseudosciaena crocea including PC、PE、PI、SM, LPC,the relative content were PC69.38%,PE4.14%,PI8.52%,SM7.24%,LPC10.72%respectively.And also compare the composition of fatty acids of Phospholipid from the egg of Pseudosciaena crocea with total lipid,soybean phospholipid.yolk phospholipid.The results showed that the soybean phospholipid,yolk phospholipid with low content of EPA and DHA.The total content of EPA and DHA in Phospholipid from the egg of Pseudosciaena crocea reach to39.1%.The total content of EPA and DHA in phospholipid is much higher than that in total lipids.5.The antioxidative activity of Phospholipid from the egg of Pseudosciaena crocea was studied,and compared with the effect of Vc including the experiment of eliminatingOH·,O2·,DPPH-and reducing capacity.The results showed that Phospholipid from the egg of Pseudosciaena crocea has obvious eliminating effective to OH·and is equal to Vc,eliminating effective to O2·DPPH·and reducing capacity is lower than Vc.In addition,antioxidation activity of Phospholipid from the egg of Pseudosciaena crocea on edible oils was studied using peroxide value(POV)by Schaal oven-storage test.The results showed that the Phospholipid from the egg of Pseudosciaena crocea are of good antioxidant effects on rapeseed oil,sesame oil,soybean oil and lard.When the Phospholipid in oils was0.1%,the antioxidative effect was as good as0.02%TBHQ.The Phospholipid added with Vc exhibit synergistic antioxidation activity in oil,but Phospholipid added with tartaric acid and citric acid only exhibit synergistic antioxidation activity in lard,it would weaken the antioxidation activity in rapeseed oil,sesame oil and soybean oil.
