The Experimental Study Of Chinese Medicine X-prescription Treatment Of Mice With Aplastic Anemia

In this paper,we use the joint application method of X-rays, cyclophosphamide（CY）and chloramphenicol （CH）, by the pathological observation of the red blood cells ofmouse peripheral hemogram, the reticulocyte count, the bone marrow nucleated cellcounts, spleen index and the bone marrow and spleen. To establish the similarity to thehuman clinical features aplastic anemia animal models, at the same time, to thetraditional Chinese medicine X-prescription treatment of AA mice dose-effectrelationship and its mechanism was discussed.The result of Aplastic anemia animal models shows:the orthogonal test methoddetermines, aftering giving the mice once body400CGy X-ray irradiation,on the4d,5d,6d intraperitoneal injection of cyclophosphamide50mg.kg^-1.d^-1, and chloramphenicol62.5mg.kg^-1.d^-1, it established the animal model of clinical manifestations of aplasticanemia successfully. the model mouse of peripheral hemogram, reticulocyte number ofbone marrow nucleated cell number and weight, and spleen index decreasedsignificantly;the spleen pathological observations show:the AA model mouse spleentissue is loose,the boundary of red and white pith blur, the number of splenic folliclesare reduced and the outline is unclear.The Bone marrow pathology observed: thenumber of hematopoietic cells was significantly reduced and almost entirely replaced byfat cells.The effect of Chinese medicine X Prescription on the treatment of aplastic anemiamice shows:①The Chinese medicine X Prescription has an important influence onaplastic anemia of mice, a significant increase in their peripheral blood as increase inreticulocytes、bone marrow nucleated cell number and spleen index; after two weeks ofthe drug, the indicators returned to normal.Conclusion:①The method Successfully established animal model of the clinicalmanifestations of aplastic anemia. Its experimental period is short, the method is simple,the results are stable, high incidence, good repeatability;②The Chinese medicine Xprescription has an important effect on aplastic anemia in mice, particularly,at the rangeof dose0.4⁰.8g.kg^-1.d^-1, the effect is significantly better than stanozolol.The Chinesemedicine X prescription is an effective program for the treatment of aplastic anemia. Its mechanism may be related to the promotion of bone marrow hematopoietic stem cellproliferation and modulation of immune function, still need to studied further.
