Buffalo industry in China has a huge potential for development, the Ministry of Agriculture also made it clear vigorously the development of the buffalo in dairy production capacity which is weak in the southern industrial zone. However, the existence of buffalo milk yield and fertility of the "double low" seriously restricted the development of the buffalo industry. Among them, the reproductive performance is particularly related to the buffalo industry for sustainable development. Our buffalo are mainly for the local buffalo with Murrah, Nili-Rafi buffalo hybrids swamp, because the domestic buffalo(number of chromosomes2n=48), Murrah and Nili-Rafi buffalo river type (number of chromosomes2n=50), hybrid offspring chromosome is polymorphic (2n=50,2n=49and2n=48), resulting in aneuploidy, lower reproductive performance, mainly obvious estrus, irregular estrus ovulation breeding pregnancy rate is low, resulting in a serious impact on the development of China’s buffalo industry. To prove hybrid buffalo estrus ovulation law and regulation and to explore methods of pregnancy diagnosis, for achieving the purpose of improving their fertility and milk yield, this experiment on the basis of preliminary studies is to further study the following three aspects:1. hybrid buffalo estrus ovulation rules:Using B ultrasound diagnosis technology to track the occurrence and development of monitoring buffalo ovarian follicles, corpus luteum, etc., as possible timely artificial insemination as achieving the purpose to improve the situation of conception rate. The results show that:hybrid buffalo estrous cycle in different seasons (summer and fall) is different, the estrous cycle was24.0±0.5days, the cycle range was23-25days(sample size n=30) in summer; the estrous cycle was22.5±1.5days, the cycle range was21-24days(sample size n=30)in fall. By the production of hybrid buffalo wave period is only a two-wave cycle. Hybrid buffalo estrous cycle in the different seasons (summer and autumn), the wave beginning and continuing time are different. The wave beginning time in wave1and wave2were2.0±0.5days and12.0±2.0days, the duration were11.5±2.5days and12.0±2.5days (sample size n=30) in summer; the wave beginning time in the wave1and wave2were1.5±0.5days and10.0±0.5days, the duration were9.0±0.5days and12.5±1.0days (sample size n=30) in fall.2.hybrid buffalo estrus ovulation regulation technology:Sixty hybrid water buffaloes were selected in this experiment study, in different seasons (summer and autumn),using PMSG+PG estrus synchronization treatment, taking the way of the neck intramuscular, PMSG doses were divided into three groups (600IU,700IU,800IU),PGc doses were based on the corpus luteum in the ovary (0.4mg or0.6mg),to compare the suitable PMSG dose. The results show that:PMSG dose was800IU, the estrus synchronization rate and conception rate were85.00%(17/20) and58.82%(10/17), significantly higher than the75.00%(15/20)and53.33%(8/15) in600IU and the75.00%(15/20) and53.33%(8/15)in700IU; the estrus synchronization rate was86.67%(26/30) in summer, significantly higher than the70.00%(21/30) in fall, the conception rate was53.85%(14/26) in summer, less than57.14%(12/21) in fall, but the difference was not significant. Thus, estrus synchronization treatment of PMSG+PGc is better and relatively stable, estrus synchronization rate is generally higher than75.00%, conception rate is generally higher than50.00%. PMSG dose of800IU/head, you can obtain relatively high estrus synchronization rate(85.00%) and conception rate(58.82%).3.hybrid buffalo pregnancy diagnostic techniques:Using B ultrasound diagnostic techniques for early pregnancy diagnosis of60hybrid buffaloes after different breeding periods, to determine the appropriate period of pregnancy diagnosis. The results showed that:B ultrasound pregnancy diagnosis20-25days after test buffaloes mating, the diagnostic accuracy rate is relatively low,70.00%(14/20), is not the best time to diagnosis; B ultrasound pregnancy diagnosis26-35days after test buffaloes mating, the diagnosis accurately rate is80.00%(16/20), pregnancy accurately rate and left accurately rate are respectively81.81%(9/11)and77.78%(7/9), is not the best time to diagnosis too; B ultrasound pregnancy diagnosis36-45days after test buffaloes mating, diagnostic accuracy rate was100%(20/20). Therefore, the best time about using of B-pregnancy diagnosis is36-45days. Only suspicious buffaloes can be retested in46-60days to confirm.
