Pleurotus cornucopiae, a common low-temperature mushroom, belongs to Eumycophyta,Basidiomycotina, Hymenomycetes, Agaricales, Pleurotaceae, Pleurotus. It contains manybioactive ingredients, such as polysaccharides, protein, and many other biological activematerials, and has a significant effect on stimulating the faction of immune system andoxidant.Zinc is one of the essential trace elements for human body, and as the most importantcomponent of many enzymes, it plays an important role in growth and development,recognition memory, maintaining the integrity of the cell and the biosynthesis of protein andnucleic acid. But there is low-level-zinc in our daily diet structure, and the zinc can not besynthesized in vivo, therefore, cultivating this mushroom will benefit food provision andrecycling of agricultural by-products.In this paper, the culture conditions of Zn-accumulated by P. cornucopiae SS-01werestudied first, then the conditions of exopolysaccharide (EPS) and intrecellular zincpolysaccharide (IZPS) in submerged culture were optimized on the basis of Plackett-Burman(PB) design and response surface methodology (RSM). The in vitro and in vivo antioxidantactivities of EPS and IZPS were evaluated. The results were as follows:(1) The biomass of P. cornucopiae SS-01reached8.82g/L,167.38%higher than thecontrol, and the rate of Zn-accumulation wss97.87%, while the optimal concentration of zincacetate was150mg/L in liquid meida.(2) The optimal conditions of EPS extraction were ethanol concentration86.97%, pH8.99and concentration multiple2.43, and the yield of EPS production reached3.96g/L. When theconditions were ethanol concentration85%, pH9and concentration multiple2, the yield ofEPS was3.69g/L, closed to the actual value (3.68g/L).(3) The conditions of IZPS extraction were water multiple20.44, extraction time2.97h and ethanol concentration84.76%, and the yield of IZPS reached5.09%. When the conditionswere water multiple20, extraction time2and ethanol concentration85%, the yield of IZPSwas5.1%, in accordance with the actual value (5.11%).(4) The polysaccharides of P. cornucopiae SS-01had in vitro significantly scavengingeffects on superoxide anion (O2-·), hydroxyl (·OH) and2,2-diphenyl-1-picrylhydrazyl (DPPH)radicals, and high reducing power. The antioxidant effects of IZPS were significant higherthan that of ESP and intracellular polysaccharide (IPS).(5) The in vivo antioxidant results showed that the EPS and IZPS of P. cornucopiae SS-01at a dosage of0-400mg/kg had significant effects on increasing the activities of superoxidedismutase (SOD), glutathione peroxidase (GSH-Px), catalase (CAT), and on reducing thecontents of malondialdehyde (MDA) and lipid peroxidation (LPO) in blood, heart, liver,spleen and kidney of mice. Moreover, the in vivo antioxidant activity of IZPS was superior tothat of IPS.(6) Three compositions, named as IZPS-1, IZPS-2, IZPS-3of IZPS and IPS-1, IPS-2,IPS-3of IPS, were obtained by the chromatography of DEAE-cellulose, respectively.The results suggest that the EPS, IPS and IZPS of P. cornucopiae SS-01had significantantioxidant activity in vitro and in vivo, which could be used as natural and potentialantioxidants.
