The positioning comparison of organic and conventional soil characteristics positioning and nutrient regulation of biological fertilizer were combined, this paper studied techno logical bottlenecks and the solutions existed in actual production of organic agricultural. By GPS-localized monitoring, indoor culture experiments, pot experiments and field trials we mainly studied:first, soil organic matter, available nutrients and microbial biomass C, enzyme activity changes in trends in different periods of organic and conventional cropping patterns; second, the temperature and water suitability of different bio-fertilizer; third, effects of bio-fertilizer on the growth of organic vegetable, cucumber and soil nutrient and soil biological characteristics.Results of GPS-localization monitor test showed, compared with initial monitoring, soil organic matter, total N, available N, P and microbial biomass C, soil enzyme activity showed a decreasing trend under continuous years of conventional cropping patterns. Soil properties were effectively improved by organic farming. Soil bulk density showed a declining trend yearly under organic cropping patterns and no mater organic8years or4years, soil bulk density were significantly decreased compared with initial monitoring. Soil pH of organic planting was significantly higher than conventional farmland. Organic farming methods for a large number of organic manure and focus on crop rotation, straw, planting green manure and other measures to improve soil fertility, soil organic matter and soil nutrient content, thereby, soil fertility were enhanced and the supply of organic crops nutrients were continued ensured. Soil microbial and enzyme activity showed a increased trend year by year in organic farming, and in the same period soil enzyme activities showed a higher lever compared with conventional cultivation.Despite that soil nutrients were effectively increased and soil environment was improved in organic farming, but organic manure application alone could not meet the nutrient needs of crops. To address the above issues, we studied the regulation of biological fertilizer on soil nutrients. Soil temperature and moisture conditions were the main factors of microbial activity and enzyme activity. We first studied the temperature and water suitability of three different bio-fertilizer with organic fertilizer application through the indoor culture experiments. Different treatment had a different temperature or water response:the optimal culture condition of treatment with organic fertilizer only was for the temperature25℃,75%moisture;"Zhonghe" bio-fertilizer combined with organic fertilizer was at the processing temperature of25℃,60%moisture; Eco-fertilizer added with organic fertilizer processing at25℃,75%moisture; The optimum conditions of Bio bio-fertilizer combined with organic fertilizer were temperature25℃, moisture60%or75%.The pot experiment results of organic vegetable showed that bio-fertilizers combined with organic fertilizer significantly increased the yields of organic vegetable, and improved roots activities and chlorophyll contents (SPAD values), but different bio-fertilizer had a different stimulation effect. Meanwhile, the combined application with bio-fertilizers had some effect on soil biological characteristics. The content of soil microbial biomass C, N by combined application of bio-fertilizers were significantly higher than CK while vegetables25days.Finally, field trials of organic farming were carried out to examine the effects of different bio-fertilizers on the growth of organic cucumber, soil enzymes activity and soil microbial biomass. In this study, four treatments consisted of control organic fertilizer only (CK),"Zhonghe" bio-fertilizer combined with organic fertilizer (ZHH), eco-fertilizer added with organic fertilizer (NST) and Bio bio-fertilizer combined with organic fertilizer (BIO). Results showed that bio-fertilizers combined with organic fertilizer significantly increased the yields of organic cucumbers, and improved roots activities and chlorophyll contents (SPAD values). Compared with CK, the yields in the ZHH treatment were increased by10.4%, and NST, BIO treatments were increased by12.4%and29.2%respectively. In the cucumber seedling stage, the differences in early flowering season and picking time, and the amount of soil microbial biomass were found between the bio-fertilizers treatments and single-organic fertilizer controls, and some differences have reached a significant level. In addition, the ZHH, NST and BIO treatments also increased soil enzyme activities of urease, acid phosphatase and soil catalase.
