Polygonatum odoratum (Mill.) Druce is one kind of the perennial Liliaceae herb. It has a good effect on reducing blood lipid, dilating coronary artery, hypoglycemic and enhancing immunity and so on. Polygonatum odoratum is significant in Pharmacological and it will be a highly economic value for resources utilization. Recently, with the further studies, people payed more and more attentions on Polygonatum odoratum and its’demand is also rapidly increasing in clinic and healthcare industry. Strict quality control and higher quality standard are appealing. Therefore, many areas take the scientific planting to satisfy people’s need for wild resources. But the cultivate seedling originated from the wile while the distribution is wildly and its germplasm is random because the climatic and enviromental conditions and so on. Also it may appear intraspecific variation; there were a phenomenon that the quality of effective components of Polygonatum odoratum was low and unsteady. The purpose of study is that compared effective components and antioxidant activity of wild Polygonatum odoratum in different places, expounded correlation between the quality of Polygonatum odoratum and the ecological factors; and analyzed the genetic diversity and relationship of diffrernt populations of wild Polygonatum odoratum from diferent distribution regions, in order to roughly illustrate the potential relationship between the quality and the genetic diversity. The main contents and results are organized as the following:1. We study on the Polysaccharide, Water-soluble component and Alcohol-soluble component and antioxidant activity (DPPH assay and·OH free radical assay) of wild Polygonatum odoratum from different origin for the first time. We can get the result that components and antioxidant activity of wild Polygonatum odoratum from different habitats had great diversity. The content of Polysaccharide is between5.75%-42.46%, the content of Water-soluble is between29.40%-63.57%, the content of Alcohol-soluble is between33.6%-61.6%. The result show that Water-soluble、Alcohol-soluble component exhibit best antioxidant ability to DPPH assay and alcohol polysaccharides has good scavenging ability to ·OH free radical.2. By the methods of stepwise regression, principal component analysis, and grey relational grade analysis, this paper study the correlations between the quality of wild Polygonatum odoratum in different places and the ecological factors. Among the test ecolo-gical factors, the mean air temperature in January and in July, mean annual precipitation, frost-free period, and soil pH and total potassium are the main factors affecting the quality of Polygonatum odoratum, with a contribution rate of99.0%. As compared with soil factors, climatic factors make more contribution to the quality of Polygonatum odoratum.Soil tatal potassium is the key factor controlling the quality, mean annual precipitation is the main decision factor, and mean air temp-erature in January is the main limiting factor.3. The genetic diversity and relationship of diffrernt populations of wild Polygonatum odoratum from diferent distribution regions is investigated using ISSR. The results show that genetic diversity in diferent geographical population of Polygonatum odoratum is plentiful, and it ability to adapt to environmental change is strong; the genetic diversity of cultivated populations is lower than wild ones; the wild and cultivated populations from the same geographical area are clustered first and the affinity relationship of Polygonatum odoratum is closer.4. We study on the DNA extraction effects from different parts of Polygonatum odoratum, this show that better quality of DNA can be extracted and all of those are suitable for ISSR analysis. The young fruit has highest polymorphism and best quality of extraction.
