Polygonatum odoratum (Mill.) Druce is one kind of the perennial Liliaceae herb. It has a goodeffect on hypoglycemic,reducing blood lipid,enhancing immunity and dilating coronary artery andso on. Polygonatum odoratum is significant in Pharmacological and it will be a highly economic valuefor resources utilization. In recent years,research on Polygonatum odoratum gradually deepening,so that in clinical and health products industry demand is also growing fast,the quality standards formedicinal Polygonatum odoratum increasingly high. The dwindling wild resources has been unable tomeettheneedsof Polygonatum odoratum.Therefore, manyareas of themore scientific andstandardizedplantingto meetdemand.The purpose of study is that assessment of germplasm resourcesof different strains of Polygonatum odoratum,including comparative morphological and agronomictraits,DNA molecular markers,The determination of the main active ingredient,in order toachieve its systematic evaluation studies,understand the quality and genetic characteristics of differentstrains of Polygonatum odoratum,provide a theoretical basis for target trait of high-yield andhigh-quality breeding work. For the protectionof biologicaldiversity and sustainable utilization forPolygonatum odoratum,promote standardizationand industrialization,have important practicalsignificanceThe main Results are as following:1.Use the remarkable analysis,grey relationalanalysis method,studies the morphological andagronomic traitsof different strainsof Polygonatum odoratum. The results show that,there aresignificant differences among theroots,stems,leaves,flowers,fruits and seeds. Setthe values for eachtrait of the ideal strain X0as:The average diameter of the rhizome branching was1.33cm,Formheight was63.00cm,leaf numberwas17,leaf area was59.00cm. Big round leaf of purple stems(B-R-A)growth vigorousthan otherstrains of Polygonatum odoratum.Each set of data which are veryclose to the ideal line,comprehensive character best.2. Application of ISSR molecular techniques performed on different strains of Polygonatumodoratum genetic diversity and relationship analysis. Extraction of genomic DNA was using theimproved CTAB method and using a new rapid genomic DNA extraction kit,and optimize theprocedure of PCR reaction. High polymorphism and abundant genetic diversity is found amongdifferent strains of Polygonatum odoratum. differentstrainsofPolygonatum odoratumis analyzedby POPGENE32software:Nei’s gene diversity (H) was0.3246±0.1675,Shannon ’s information index(I) was0.4807±0.2270,the percentage of polymorphic loci (PPL) was86.72%,UPGMA dendrogramto eight different strains clustered into two broad categories Yuzhu also reveals their morphologicalcharacteristics Yuzhu genetic characteristics are significantly correlated,and their morphologicalcharacteristics and genetic characteristics of their habitat was no significant correlationamong.UPGMA clusteringdendrogram ofeightdifferent strains of Polygonatum odoratumclusteredinto two categories, alsoreveals thesignificant correlation among morphological characteristicsandgeneticcharacteristicsof Polygonatum odoratum,andits morphologicaland geneticcharacteristicandgrowth have no significant correlation.3. Determined the polysaccharide of different strains, different harvest time,different growthyears of Polygonatum odoratum. The study shows that the optimumharvest periodof Polygonatumodoratum polysaccharide content the2010edition of《Pharmacopoeia of The People’s Republic ofChina》of not less than6%,can be used as medicine.The best harvest period was late September ofmost of Polygonatum odoratum,only a handful of Polygonatum odoratum was late August. JiZhuNo.1although the yield per square meter and the dry weight was not obviously better than the othertest Polygonatum odoratum, but its drying rate is better than other strains, and its totalpolysaccharides are also higher than other strains,the total polysaccharide JiZhu No.1(JZYH) is thehighest,was15.49%,the followed by Small leaves purple stems(S-L-A),was15.03%.
