The seed biological characteristics, ecological condition of germination, pregermination method and storage physiology of Mytilaria laosensis and Zenia insignis seeds were studied. The results were shown as follow:1. The seedcoat of Mytilaria laosensis seeds had an excellent character of water absorption. The differentiation of seed embryo was completed. The reason of causing difficult germination might be the existence of inhibitors or low content of growth hormoner. The best treatment was300mg·L-1A3for48hours+cold stratification for45days, which could raise germination percentage of Mytilaria laosensis seed to84%. The soluble sugar content, POD activity and amylase activity were firstly decreasing and then increasing, but the soluble protein content was firstly increasing and then decreasing during the process of overcoming dormancy.2. Results showed that Zenia insignis seeds didn’t have physiological dormancy. The main reason of seed dormancy was permeability of water. The best treatment was98%sulphuric acid for20minutes or75℃constant temperature water bath for1minutes, which could respectively raise germination percentage to97%nd93%, and decreased the rate of hard seeds and mortality in the maximal degree.3. The experimental results of the effect of ecological factors on germination showed that the optimum germination temperature was25~30℃(constant temperature) for Mytilaria laosensis and Zenia insignis seeds. Two kinds of seeds were insensitive to light period, but the light period had effect on germination rate, uniformity and radicle growth. The Germination rate and uniformity of Mytilaria laosensis seeds were higher and radicle growed sturdily in the condition of12hours light/12hours dark. The Germination rate and uniformity of Zenia insignis seeds were higher and radicle growed sturdily in the condition of continuous light condition.4. Germination, germination energy, germination index, POD activity, SOD activity, CAT activity, soluble sugar content and soluble protein content of Mytilaria laosensis and Zenia insignis seeds decreased while relative conductivity and MDA content increased with the prolong of storage time in different storage condition(room temperature,4℃,—20℃). Principal component analysis indicated that there was high correlativity between seed germination and test physiological indices. It showed that seed aging was the result of a series of physiological change during storage. The storage condition of—20℃was the most favorable for keeping aging and activity of Mytilaria laosensis seeds. Secondly, the condition of4℃was better. The storage condition of4℃was the most favorable for keeping aging and activity of Zenia insignis seeds. Secondly, the condition of—20℃was better. These results showed that low temperature was beneficial to keep activity of two seeds.
