To reveal the influence of probiotics on oxidation and anti-oxidation of chickens which were vaccinated by IBD vaccine, challenged with virulent IBDV, the methods of histochemistry staining and immunohistochemistry were used to detect the changes of T-SOD, MDA and NO content in serum and some organs, such as heart, liver, spleen, kidney and bursal, and the changes of histopathology were also observed in order to reveal the damage of oxidation. The different types of SOD in organs were detected by PCR. This experiments could provide the scientifitic evidence for the development of probiotics. The results were as follows:1. After one-day-old chickens fed with probiotics, the content of T-SOD in serum and organs was much stronger than that of controls (P<0.05or P<0.01), after challenged with virulent IBDV, the content of T-SOD in serum and organs was much stronger than that of controls and chickens which were just vaccinated by IBD vaccine without probiotics fed (P<0.01). These results indicated that the anti-oxidation enzyme was expressed stronger in chickens fed with probiotics after challenged with virulent IBDV, and, the function of anti-oxidation was inhanced remarkably.2. The content of MDA in serum and organs was much more lower than that of controls and chickens which were just vaccinated by IBD vaccine without probiotics fed (P<0.05or P<0.01). By observing the changes of histopathology, the damage of probiotics fed chickens organs was much more lighter than that in controls and chickens which were just vaccinated by IBD vaccine without probiotics fed. The bursal has the fiber hyperplasia, including reticular fiber, collagenous fiber and elastic fiber which was the least fiber content. These results indicated that the probiotics fed chickens could be better protected from damaging after challenged with virulent IBDV than that in controls and chickens which were just vaccinated by IBD vaccine without probiotics.3. The content of NO in serum and organs including the content of iNOS in organs was lower than that of controls (P<0.01), neverthelessly, it was higher than that in chickens which were just vaccinated by IBD vaccine without probiotics fed (P<0.05or P<0.01). The results indicated that probiotics could stimulate the macrophage proliferation which could result in upregulating the content of iNOS in organs. NO has the anti-virus activity and cytotoxic activity, the latter was more obvious in virulent IBDV infected chickens.4. Probiotics Ⅰ group chickens with Lactobacillus primarily, the protection of anti-oxidation was better than probiotics Ⅱ group chickens with Bacillus primarily.
