Chimonanthus grammatus,the unique rare plant germplasm resource in Jiangxiprovince,belongs to Chimonanthus Lindl. of Calyzanthaceae. C. grammatus is rich involatile oils, alkaloids, flavonoids, and sesquiterpenes and other natural chemicalcomposition, which have high potential and great application prospects in the fields ofmedicine, aromatic oil, etc. However, the researchs about C. grammatus focuse on itsbiological characteristics and community structure now,while study on the chemicalcomposition and pharmacological activity of the plant is still very lacking, and thepharmacological mechanism is not yet clear. In this paper,with activity as a guide,westudy the C. grammatus leaves’ chemical composition, and analyse the antibacterialactivity, antioxidant activity and inhibition activity on acetylcholinesterase of thecompounds with biological activity qualitatively and quantitatively,in the purpose ofproviding basic data and scientific basis for better development and utilization of thisvaluable species.In this study, by ethanol extraction method, we got C. grammatus concentratedleaf extract,and obtained five kinds of crude extract by a combination of acid-baseprocessing and extraction of polarity: the CGⅠ-chloroform phase, the CGII-alkalinephase, CGIII-ethyl acetate phase, CGIV-acidic water phase, the CGⅤ-ethyl acetatephase.By some screening techniques, we determined the CGⅠ, CGIII, CGⅤ as theactive component. A total of13kinds of compounds were isolated by the separationmeans such as repeated silica gel column chromatography and preparative TLCchromatography.The structures were analysed by NMR spectroscopy, UVspectroscopy, mass spectrometry (MS) analysis and infrared spectroscopy analysisand nine compounds were identified: l-Chimonanthine(2), β-Sitosterol(3),Stigmasterol(4), Daucosterol(5), Prenylated pHenolics(8),(E)-4-(4,8-dimethylnona-3,7-dienyl)furan-2(5H)-one(9), Diisobutyl phthalate(10),Quercetin(11), Scopoletin(12). Four kinds of compounds, Prenylated pHenolics(8),(E)-4-(4,8-dimethylnona-3,7-dienyl)furan-2(5H)-one(9), Diisobutyl phthalate(10),Scopoletin(12) were isolated in this genus for the first time.Monomer compounds’ activities were detected:compound8, compound9andcompound10show good antibacterial activity on Bacillus subtilis and Escherichiacoil; compound1, compound9, compound10has good antioxidant activity;compound1shows good inhibition effects on AChE activity. MIC/MBC of theactive compounds were detected on E. coli and Bacillus subtilis.Meanwhile the DPPHradical scavenging activity of the active compounds were detected.
