The idea of using Chinese traditional medicine as wood preservative is aim at environmental problems of existing preservatives. The herbal medicines can be said to be an absolute environmental protection materials, most of them have anti-bacterial, anti-microbial and anti-fungal effect. So choose some Chinese traditional medicine to prepare wood preservatives has certain theory foundation. The purpose of this research is to study the anti-fungi mechanism of different formulations of traditional Chinese medicine on the wood rot (white-rot fungus, brown rot fungi). We used biological and chemical methods, electron microscopy chemical cell technology, growth curve, two-dimensional electrophoresis, mass spectrometry, oxygen measurement system and so on. In order to detect and analyze the anti-fungal act and mechanism of Chinese medicine on the wood-rot fungi, provide the theoretical basis and data to support the further exploitation and utilization of a new preservative:Chinese traditional medicine, then solve fundamentally the environmental problems of wood preservatives, at the same time, we can find a new way of wood preservation.We used six kinds of antibacterial Chinese herbal pharmaceutics as wood preservative:Cnidium monnieri, Sophora flavescens, Coptis, forsythia, Stemona and Kochia scoparia, and wood decaying fungus(Ceriporiopsis Subvermispora and Monilinia fructicola) as experimental subject. This research studied on the antibacterial mechanism of preventing wood from decaying by analyzing the antibacterial property of Chinese herbal pharmaceutics, the toxicity of Chinese herbal pharmaceutics to wood decaying fungus’s morphology, the influence on the growth cycle of decay fungi, the influence pharmaceutics to wood decaying fungus’s metabolism and protein. The results showed that the Chinese herbs chose all have the antibacterial property. Turbidimetric method draw out the growth curve of decay fungi with different Chinese herbal medicine, and it is mainly to inhibit decay fungi growth in the growth logarithmic phase to affect its growth cycle. The observation of SEM shows that Chinese herb has inhibitory action to the morphology growth and development of the ceriporiopsis subvermispora obviously. The mycelium morphology has changed greatly, while the growth and development are malformed. The test conclusion of two dimentional electrophoresis indicates that Chinese herbal pharmaceutics hinders the formation of some proteins and enzymes and inhibits the formation of DNA by influencing fungus controls the expression of related DNA biosynthesis gene, which effect on the formation of certain protein which is necessary for thallus cell division, hinder gene expression of some protein and enzyme related on the thallus’s normal growth, and make the tallus die with its disaggregation. The research on oxygen electrode method shows that Chinese herbal pharmaceutics can produce certain influence to ceriporiopsis subvermispora’s respiratory metabolism. Through the studies in the project, the wood preservative mechanism of Chinese herbal pharmaceutics is mainly influent Chinese herbal pharmaceutics to wood decaying fungus’s respiratory metabolism and the expression of related DNA biosynthesis gene, which make the thallus grow and develop abnormally, and disaggregate to death to preservation. From the long-term development perspective, the test is to make full use of the existing plant resources and develop the green plants source preservatives which harmony with the biological and environmental, provide a theoretical basis and technical support for the development of new wood preservatives, and also provide a new approach for the comprehensive development and utilization of the Chinese herbal plant resources.
