Many ingredients of Chinese herbal medicine can enhance animal immune function, inwhich polysaccharide is the main immune active compound of Chinese herbal medicineimmunopotentiator. At present, the extracting technology and methods of polysaccharideare numerous, such as solvent extraction, acid extraction, alkali extraction, microwave andultrasonic assisted extraction method etc. The different extracting technology can producedifferent extraction rate. So, it has realistic meaning to explore the optimum technologicalcondition for extracting high yield polysaccharide from Chinese herbal medicine.The response surface methodology (RSM) was adopted in this experiment to exploreoptimal conditions for extracting polysaccharide from compound Chinese herbal medicine.The polysaccharide extracted under optimal conditions was diluted into three gradients forthe study of immune-regulating effect and mechanism in chicken.300chicken (1day old)were randomly divided into5groups of60chicken each, matched by weight. TheGroupⅠwas as the control group, the GroupⅡ was a0.04%APS group, the Group Ⅲ wasa0.02%polysaccharide group, the Group Ⅳ was a0.04%polysaccharide group, and theGroup Ⅴ was a0.08%polysaccharide group. The experiment was conducted for42days.10chicken in each group were selected to collect blood samples and slaughter on day14,21,28,35and42for determining the specific immunity, nonspecific immunity, immuneorgans index and mucous membrane immunity.The results were as follows:1. The RSM for extracting polysaccharide from compound-Chinese herbal medicineThe optimal conditions for extracting polysaccharide from compound-Chinese herbalmedicine by the ultrasonic-assisted extracting method were: the ratio of solid to liquid was1:20, the extraction time was40min, the temperature was80℃, the power of ultrasoundwas360W, and the extracting rate of polysaccharide was17.4735%.2. The effect of polysaccharide from compound-Chinese herbal medicine on specificimmunity in chickenThe antibody titer of ND, lymphocytes transformation activities and protein contentswere detected. The results showed that, compared with the control group and0.04%APSgroup, the ND antibody titers, lymphocytes transformation activities and protein contentsof chicken in the0.04%polysaccharide group were significantly different on day21afterimmunized (P﹤0.01). The results indicated that the polysaccharide fromcompound-Chinese herbal medicine could significantly enhance the speed of the antibody response, stimulate animals to produce protection antibody earlier, and increase cellularimmunity of chicken in the early growth stage.3. The effect of polysaccharide from compound-Chinese herbal medicine onnonspecific immunity in chickenThe results showed that, compared with the control group and0.04%APS group,the cytophagic index and the contents of serum lysozyme of chicken in0.04%polysaccharide group were significantly different on day28after immunized (P﹤0.01orP﹤0.05). The results indicated that the polysaccharide from compound Chinese herbalmedicine could increase the serum lysozyme contents and macrophage activities of thechicken in the early stage, improve the capacity of nonspecific immune.4. The effect of polysaccharide from compound-Chinese herbal medicine on immuneorgan indexes in chickenThe results showed that, compared with the control group and0.04%APS group,the indexes of thymus, spleen and cloacal bursa of the chicken in0.04%polysaccharidegroup and0.08%polysaccharide group were significantly different on day28and35afterimmunized (P﹤0.01). The results indicated that the polysaccharide from compoundChinese herbal medicine could promote the growth of immune organs and enhance theimmunity of chicken.5. The effect of polysaccharide from compound Chinese herbal medicine on mucosalimmune system in chicken.The results showed that, compared with the control group, the mucous membraneantibody levels of the chicken in0.04%polysaccharide group were significantly differenton day21,28and35after immunized (P﹤0.01), but not different compared with the0.04%APS group (P﹥0.05). The results indicated that the polysaccharide from compoundChinese herbal medicine could induce mucosal immune reactions, improve the localimmunity effectively.The experiment confirmed that the immune-regulating effect of polysaccharide fromcompound Chinese herbal medicine in the0.04%of polysaccharide group of chicken wassimilar to that in the0.08%polysaccharide group of chicken. So it was concluded thatthere was an optimal dose of the polysaccharide in enhancing immunity of chicken.Therefore, the0.04%polysaccharide was defined as the optimal dose according to theeconomic viewpoint.
