Mal-environment and stress are the main factors of immunity deficiency and blight spread in intensive nursery. Therefore, to enhance immunity is very important for preventing and controlling ailment.Immunopotentiating agent has broad development and application prospect mainly because its ability to recover and increase body fluid immunity and cellular immunity when animal’s immune function is low. At present, immunopotentiating agent applicated in clinical medicine include biological, chemical, nutrition and bacterial agent, most of them are good for its active adjuvant, but there are also some drawbacks, such as drug residue and allergic reaction. So searching for safety, reliable, no medicine residual and rapid immunopotentiating agent become urgent problem to be solved in livestock production currently.As natural drug, Chinese herbal medicine has become primary choice as immunopotentiating agent because of its favourable characteristics, such as rich resources, low cost, small side effects, no residue and uneasy resistance etc. Study showed that many ingredients of Chinese herbal medicine could affect body’s immunity regulate function, in which polysaccharide was the main immune enhancement active compounds.Compared with fat-soluble chemical composition, polysaccharide has been ignored in chemistry and pharmacological research for many years, which has been eliminated as impurities. One reason is it’s role remain unclear, the other reason is due to it’s separation difficult. With the development of science and technology, Chinese traditional medicine polysaccharides extract method has been developed to cellulose enzymatic, ultrasonic-assisted extraction and ultrafiltration method from its original water decoction method. The extraction ratio of polysaccharide are various considerably by different extraction technology and method. So, it has realistic meaning to grope the optimum technological condition for extracting high yield polysaccharide from chinese herbal medicine.The orthogonal design was used in this paper to explore optimal conditions for extracting polysaccharide from compound-Chinese herbal medicine which used the method of ultrasonic-assisted extraction. Compared with the compound decoction of compound-Chinese herbal medicine group, the polysaccharide which extracted under optimal condition were diluted into three gradients and used for the study of immunity regulate function and mechanism in mice. The motive is to explore good and cheap Chinese herbal medicine immune enhancement agent and provide dependable basis for its development and utilization.Two hundred mice were divided into five groups randomly, saline group(group A), compound decoction of Chinese traditional medicine group(group B), low concentration of polysaccharide group(group C), medium concentration of polysaccharide group(group D) and high concentration of polysaccharide group(group E). Each group contained40mice and every mouse was fed0.3mL liquid successively for7d. They were killed on the8d, mice’s immune organ’s weight, function of cellular immunity, body fluid immunity and mononuclear phagocytic were determined.The results were as follows:The optimal conditions for extracting polysaccharide from compound-Chinese herbal medicine which used the method of ultrasonic-assisted extraction were: the number of ultrasound is36, the power of ultrasound is800W, and the ratio of solid to liquid is1:30. The extraction rate of polysaccharide is6.602%and the saccharinity is71.293%.Compared with saline group and compound decoction of compound-Chinese herbal medicine group, the data of body fluid immunity, cellular immunity and so on were increased significantly in polysaccharide for compound-Chinese herbal medicine group. So we can conclude that polysaccharide for Chinese herbal has significant effect on immune system of mice.The function of increasing the mice’s immunity in high polysaccharide group was similar to that in medium polysaccharide group. So it can be concluded that there is an optimal concentration of the polysaccharide for enhancing mice immunity fuction. From an economic point of view, the medium polysaccharide dose is determined as the optimal dose. Acute Toxicity Tests showed that compound traditional Chinese medicine had very low toxicity, so it’s clinical application will be safe. Therefore, it has great prospects for development and application of the polysaccharide which extractd from compound-Chinese herbal medicine by ultrasonic-assisted extraction method.
