Mastitis is one of the important diseases of serious harm to the dairy industry, and also one of the diseases caused huge economic losses to the dairy industry. Staphylococcus aureus is the main pathogens of mastitis, can cause cows breast tissue inflammation, fibrosis, and seriously affect the quality of the milk and the milk production of years.The adhesin is the most important virulence factor in Staphylococcus aureus early infection. Thus blocking the initial stages of Staphylococcus aureus infection, may be the most effective strategy to preventing infections. In this study, the popular Xinjiang three different areas of Staphylococcus aureus fibronectin-binding protein (FnBPA) object for the study and analysis of their genetic characteristics. Investigate whether the FnBPA-A genetic polymorphism affect immunological properties the the immunogen to FnBPA. In this study, Staphylococcus aureus FnBPA functional genes were amplified by PCR from Xinjiang three different regions, and sequenced. The sequencing results showed that the target gene had been cloned. The results of homology comparison indicated that the Xinjiang region FnBPA-A nucleotide sequence homology is from76.6%to100%,amino acid sequence homology is from71.4%to100%. the phylogenetic tree is divided into three main branches, shows that different regions of Xinjiang the Staphylococcus aureus FnBPA-A sequence has some differences.FnBPA-A gene were connect to the eukaryotic expression vector pVAXl, FnBPA-A recombinant plasmid were constructed sucessfully. The indirect immunofluorescence results showed that the target protein can be successful expression in293T cells. The animal experiments showed that the antibodies produced after immunization with different characteristics.The results of this study will provide experimental basis for future rational design Staphylococcus aureus vaccine for the region and the effective control of cow mastits.
