ObjectiveTo use RTFQ-PCR to explore whether there are differences in apoptotic genes, the recoveryof the function of the injuried spinal cord,and the pathological changes between the ASCI groupand the MP treatment group.Methods1. To build the ASCI models,the improved Allen’s weight drop method is taken. All animals areselected randomly and divided into these three groups. Rivlin loxotic plate scores and BBBScoring system are chosed to evaluate the function of the spine.The score are collected at sixtime points after the ASCI, which are the8hour,24hour,72hour,7day,14day and28day,andthese scores are analyzed among groups;2. Animal models are selected randomly and sacrificed according to the6time points to get theHE staining to analysis the pathological differences;3. All rabbits were sacrificed at each time point of8hour,24hour,72hour,7day,14day and28day, using RTFQ-PCR to detect the quility of bcl-2and bax gene,analyze the differencesbetween group C and T,and explore the possible mechanisms of ASCI treated by MP;4. Using RT-PCR to detect the expressed quility of cytc and caspass-9between groupC and T,and explore the possible mechanisms of ASCI treated by MP.Results1. Loxotic plate values and BBB scores increased in the group C and group T according to thetime order of8hour,24hour,72hour,7day,14day and28day. For the The Loxotic platevalues,statistical differences starts at the time point of7day(P<0.05).But for the BBB scores,statistical differences start in the time point of14day(P<0.05);2. No pathological changes by HE staining are found in Group S. Between group C and T,pathological changes could be found in the time point of7day,14day and28day.there are nosignificent changes between group C and T in the time point of8hour,24hour and72hour;3. Using RTFQ-PCR to detect the expression of the bcl-2gene in group C and T,in all the timepoint,the expressions of bcl-2are higher in group T,and there are statistic differences(P<0.05);4. Using RTFQ-PCR to detect the expression of the bax gene in group C and T,in all the timepoint,the expressions of bax are higher in group C,and there are statistic differences(P<0.05),in the24h72h and7d,C group express the same like T group; 5. Using RTFQ-PCR to detect the expression of the Cytc and Caspass-9gene in group C and T,inall the time point, the expressions of Cytc and Caspass-9are higher in group C than groupT,and there are statistical differences(P<0.05).And from the line of the Cytc and Caspass-9ofthe group T, there are some relationship between them.Conclusion1. The ASCI models recover better when treated by MP than the models that were not treated byMP;2. MP have the effect of reducing the exudation of the red blood cells and inflammatorycells,which is related with the function of the protection of the injuried spinal cord;3. bcl-2expressed higher in the MP treztment group than in the non-MP treatment group;4. The expression of Cytc and Caspass-9are lower in the MP treatment group.
