Objective: To study the mRNA level and the expression of p27kipland Jab1protein inovarian epithelial cancer and its significance.Methods: Detect the mRNA content and protein expression of p27kip1and Jab1ofhuman in44cases of epithelial ovarian cancer tissue,30cases of benign ovarian tumortissue and30cases of normal ovarian tissue by Reverse Transcriptase Polymerase ChainReaction (RT-PCR) semi-quantitative method and immunohistochemical staining,respectively. Analyze the relationship between p27kip1and Jab1in epithelial ovarian cancertissues, and the relationship between p27kip1and Jab1with clinical pathological featuressuch as clinical stage and cell differentiation of epithelial ovarian cancer.Results:1. The mean mRNA level of p27kip1was0.2091in epithelial ovarian cancer tissue,0.5423in benign ovarian tumor tissue and0.6417in normal ovarian tissue. The positiverate of p27kiplprotein expression was34.09%(15/44) in epithelial ovarian cancer tissue,70%(21/30) in benign ovarian tumor tissue and80%(24/30) in normal ovarian tissue.There were no statistical difference in benign ovarian tumor tissue and normal ovariantissue (P>0.05),but the mRNA level of p27kip1and the positive rate of p27kip1protein wassignificantly decreased in epithelial ovarian cancer tissue compared to that in the othergroups (P<0.05).2. The mean mRNA level of Jab1was0.9824in epithelial ovarian cancer tissue,0.5711in benign ovarian tumor tissue and0.5280in normal ovarian tissue. The positiverate of Jabl protein expression was84.09%(37/44) in epithelial ovarian cancer tissue,53.33%(16/30) in benign ovarian tumor tissue and36.67%(11/30) in normal ovariantissue. There were no statistical difference in benign ovarian tumor tissue and normal ovarian tissue (P>0.05), but the mRNA level of Jab1and the positive rate of Jab1proteinwas significantly increased in epithelial ovarian cancer tissue compared to that in the othergroups (P<0.05).3. The mRNA level and positive expression rates of p27kipland Jab1proteins inovarian epithelial cancer was correlated with clinical stage and pathological grade (P<0.05), but have no significant correlation with the age, the size of solid tumors and whetherthere is lymph node metastasis (P>0.05).4. In ovarian epithelial cancer, p27kipland Jab1level of mRNA was negativelycorrelated (r=-0.949, P<0.05); and protein expression of p27kipland Jab1was negativelycorrelated (r=-0.474, P<0.05).Conclusions:1. Reduced expression of p27kipland higher expression of Jab1may play importantroles in the incidence and development of ovarian epithelial cancer.2. The expression of p27kipland Jab1was negatively correlated in the epithelialovarian cancer tissue.3. The detection of p27kipland Jab1in mRNA and protein expression levels may beconsidered as important indicators of determine the type of epithelial ovarian tumors andearly diagnosis and treatment of ovarian cancer patients.
