Diabetes and diabetes chronic complication is a serious disease for human health. At present, cassia seed on diabetes complications is mainly on the compound recipe treatment. In order to study the mechanism of traditional Chinese medicine, this article mainly studies on the single tradition Chinese medicine herb cassia seed on the activity of anti-diabetic complication and research the mechanism on oxidative stress.The content and the main results of the study are as follows:1、By researching the compound recipe on the treatment of diabetic complications for 30 years and analysis the frequency of the herb, screening of the best activity of 74 kinds of the Chinese herbs. Through the index of fasting blood glucose values, the activity of AR and SOD, the content of MDA and the kidney/body weight to select cassia seed in the animal model of diabetic induced by alloxan. So change the study on compound recipe to the single tradition Chinese medicine herb.2、After the extraction and isolation from semen cassiae, screening the more activity extractions using the experiment of HepG2 cells and the inhibition of the ROS. The results showed ethyl acetate is the best activity extraction. Isolation the ethyl acetate and get four compound, chrysophanol, physcione, emodin and rhein. Using the same method to compare the activity of the four compounds and found emodin and ehein is the activity compound.3、Study on the activity extractions and compounds on the diabetes mice model. To evaluate the activity of the anti-diabetes complications by detecting the body weight, diet and water intake, fasting blood glucose values, activity of AR and SOD and the content of MDA. The result showed that both the ehein and emodin are the activity compounds from ethyl acetate, and ehein is much better than emodin which can improve the body weight, diet and water intake, and the oxidative stress in the diabetes mice.4、Research the mechanism on oxidative stress of the activity component. ROS is the core mechanism of the diabetic complication. The main source of ROS is the produce of the mitochondrial respiratory chain. The result showed that ehein and emodin can suppress the ROS in the mitochondrial. It indicate that the inhibition of the reactive oxygen species generation in the mitochondrial may be one of the mechanisms of the treatment of diabetic complication.
