OBJECTIVE To prepare the Semen Cassiae prescription Transfersome and to investigateits influence on mRNA expression of adipose triglyceride lipase (ATGL) and G0/G1switchgene2(G0S2) in adipose tissue of diet-induced obese rats.METHODS1. Semen Cassiae prescription Transfersome was prepared byfilm-ultrasonic dispersion method. The effects of the ratio of different components in thetransfersome products on encapsulation efficiency were investigated. The rates ofenvelopment of Semen Cassiae prescription transfersome were determined by ultravioletspectrophotometry. The particle size and Zeta potential of Semen Cassiae prescriptiontransfersome were evaluated by Zetasizer analyzer.2.60male Sprague-Dawley rats weredivided into groups. After8-week treatment, body weight, wet weight of visceral fat andpercentage of body fat (PBF) were measured. The levels of fasting blood glucose, serumlipids were assessed. The adipose tissue section was stained with Hematoxylin-Eosin, andthe cellular diameter and quantity of adipocytes were evaluated by light microscopy. ThemRNA expression of ATGL and G0S2from the peri-renal fat tissue was determined byRT-PCR.RESULTS1. The appropriate formulation was deoxycholic acid sodium salt vs.phospholipids1:8, cholesterol vs. phospholipids1:3, vitamin E vs. phospholipids1:20,Semen Cassiae prescription vs. phospholipids1:6. Zeta potential was-15.11mV and theparticle size was292.2nm. The mean encapsulation efficiency was (69.350.25)%.2.Compared with the obese model group, treatment with Semen Cassiae prescriptiontransfersome resulted in significantly lower body weight, wet weight of visceral fat, PBFand mRNA expression of G0S2from peri-renal fat tissue (all P<0.05), and significantlyhigher level of high-density lipoprotein cholesterol (HDL-C)(P<0.05), diameter ofadipocytes, and mRNA expression of ATGL from peri-renal fat tissue (P<0.05).CONCLUSION The preparation method is simple and reasonable. The Semen Cassiaeprescription transfersome with negative electricity was small and uniform particle size. Andit had high encapsulation efficiency and stability. Semen Cassiae prescription transfersomecould reduce body weight and adipocyte size; and the effect was associated with theup-regulation of ATGL and down-regulation of G0S2expression in the adipose tissue andimprovement of insulin sensitivity.
