Objective To observe the effect of recombinant TGF-β IL-6and IL-23on differentiation and proliferation of Thl7cells in murin viral myocarditis induced by coxsackie virus B3, and further investigate the role of these cytokines act on differentiated Th17cells, which provide new ideas to Th17cells as a target for the treatment of VMC.Methods VMC model was induced in6weeks old male Balb/c mice by Coxsackie Virus B3peritoneal injection. Murin myocardial were prepared one week later for histopathology observation. Murin splenocytes were also prepared, CD4+T cells were isolated by immunomagnetic beads then cultured and divided into control group; TGF-β+IL-6group; TGF-β+IL-6+IL-23group in vitro.5days later, the frequencies of Th17subsets in CD4+T cell was evaluated by flow cytometric analysis. The expression of IL-17mRNA was analyzed by RT-PCR. And the concentration of IL-17in the culture supernatant was assayed by ELISA.Results (1) Myocardial tissue of1week VMC mice has appeared focal inflammatory cells infiltrating performance.(2) Compare with the control group, the frequencies of Th17cells increased slightly in TGF-β+IL-6group ([1.64±0.43]%vs[1.96±0.35]%respectively), the expression of IL-17mRNA and the concentration of IL-17in the culture supernatant also higher than control ([0.47±0.15] vs [0.34±0.07] and [19.01±4.02] pg/mL vs [24.57±4.10] pg/mL respectivly), but the statistical difference wasn’t significant (P>0.05).(3)In TGF-β+IL-6+IL-23group, the frequencies of Th17cells increased obviously (4.08±0.77)%, the expression of IL-17mRNA and the concentration of IL-17in the culture supernatant also augmented apparently ([0.75±0.07] and [61.19±15.18] pg/mL respectively), which shows statistical significance than the other two groups (P<0.05)Conclusion(1) Exogenous TGF-(3in combination of IL-6stimulation have no effect on the differentiation and proliferation of spleen differentiated Thl7cells in VMC mice.(2) Exogenous IL-23increase the population of Th17cells and up-regulate the level of IL-17significantly, which indicate that IL-23plays an important part in the proliferation and differentiation of Th17cells, and may directly or indirectly affect the inflammatory response of VMC...
