OBJECTIVE To explore the effect of IL-17A knockout on anti-adeninenucleotide translocator (ANT) autoantibodies, anti-β-myosin heavy chain(β-MHC) autoantibodies and anti-cardiac L-type calcium channel (CACH2)autoantibodies in mice with viral myocarditis.METHODS Male wild-type (WT) and IL-17A-deficient (IL-17A-/-)mice on the BALB/c background were intraperitoneally (i.p) injected withcoxsackie virus B3(CVB3) for establishing VMC models (VMC-WT groupand VMC-IL-17A-/-group). Meanwhile, a control group (WT group) of WTmice were established and treated with phosphate buffered saline i.p. Paraffinsections of cardiac tissues were made14days after CVB3injection.Myocardial histopathologic changes were evaluated by hematoxylin-eosinstaining. The cardiac indexes and the cardiac pathological scores werecalculated. The levels of anti-ANT autoantibodies, anti-β-MHC autoantibodiesand anti-CACH2autoantibodies from blood serum were measured byenzyme-linked immuno sorbent assay (ELISA). RESULTS Compared with WT group, the cardiac indexes and thecardiac pathological scores were significantly increased in VMC-WT group(P<0.05, P<0.01), the levels of anti-ANT autoantibodies, anti-β-MHCautoantibodies were significantly increased in VMC-WT group (P<0.05), whilethe concentration of anti-CACH2autoantibodies shown no significantdifference between WT and VMC-WT group (P>0.05). Compared withVMC-WT group, the cardiac indexes and the cardiac pathological scores weresignificantly increased in VMC-IL-17A-/-group (P<0.01, P<0.01), the level ofanti-ANT autoantibodies was reduced in VMC-IL-17A-/-group (P<0.05), whilethe levels of anti-β-MHC autoantibodies and anti-CACH2autoantibodiesshowed no significant difference between them (P>0.05).CONCLUSION1. IL-17A knockout can restrain the inflammation ofheart in VMC mice.2. The anti-ANT autoantibodies and anti-β-MHCautoantibodies level of serum in VMC mice is obviously increased.3. IL-17Acontribute to the secretion of anti-ANT autoantibodies of VMC mice.4. IL-17Ahas no effect on the secretion of anti-β-MHC autoantibodies and anti-CACH2autoantibodies in VMC mice.
