Objective:To study the relationship between family constellation of hepatocellular carcinoma (HCC) and the polymorphism of glutathione S-transferase Ml and Tl in Fusui county of Guangxi province.Methods:Peripheral blood samples were collected from76members of21HCC clustering families and68members of21control families, among Zhuang population, in Fusui county of Guangxi province. The area had been with high incidence rate of HCC. Genotypes of glutathione S-transferase Ml and T1were identified by multiplex polymerase chain reaction. Combining trial results with clinical materials were analysed statistically by case-control study.Results:1. The differences of frequencies of GSTM1or GSTT1null genotype and GSTM1and GSTT1combined null genotype were not significant between the first-degree relatives and probands of HCC 2.The frequencies of GSTM1null genotype in the probands of HCC and controls were71.4%and36.8%respectively(x2=7.791,P=0.005).The frequencies of GSTT1null genotype in the probands of HCC and controls were47.6%and19.1%respectively(x2=6.801,P=0.009).The prevalence of GSTM1and GSTT1combined null genotype in the probands of HCC was33.3%as compared with2.9%in controls,showing a remarkable difference between them (x2=13.132,p=0.000).3.The frequencies of GSTM1null genotype in the family constellation of HCC group and controls were67.1%and36.8%respectively(x2=13.256, P=0.000).The frequencies of GSTT1null genotype in the family constellation of HCC group and controls were40.8%and19.1%respectively(x2=7.944, P=0.005).The prevalence of GSTM1and GSTT1combined null genotype in family constellation of HCC was31.6%as compared with2.9%in controls. showing a remarkable difference between them(x2=18.005,p=0.000).4.Take the GSTM1and GSTT1combined present genotype as the reference,the odds ratio calculated between the family constellation of HCC group and controls about GSTM1(-)/T1(-).GSTM1(+)/T1(-).GSTM1(-)/T1(+) were3.092.0.210.0.102respectively.Conclusions:1.The relationship had existed between familial clustering of HCC and the f-requencies of polymorphism of GSTM1and GSTT1.2.The gene deficiencies both GSTM1and GSTT1were positively correlated with the liver cancer occurrence,and there was a synergistic effect in hepatocarcinogenesis.
