Objective:To Observe the effect of exogenous matrix metalloproteinases (MMP-2) and matrix metalloproteinase inhibitors (TIMP-2) to wound healing and the healing scar within the medical reference range of small doses.Methods:Selected34SD rats and made the model of rat back skin wound healing,rats would be randomly divided into the MMP-2groups,TIMP-2groups and phosphate buffered saline (PBS) control group,and given these three reagents and compared. Postoperative we observed the wound healing, granulation tissue growth in daily, and recorded wound healing time, measured and recorded wound healing scar size.Postoperative we drawn some samples, and made histological observation,observed the collagen morphology, detected a-smooth muscle actin (alpha-SMA) expression by the chemical technology and image analysis.Results:①Thea-smooth muscle actin of fiber cells in the TIMP-2group was significantly higher than the PBS and MMP-2group,and it was significantly difference between the PBS and MMP-2group (P<0.05);②Compared with the others groups,the wound average healing time in the TEVIP-2group was significantly reduced (P<0.05),and the PBS group was shorter than the MMP-2group, there was statistically significant (P<0.05)③The scar thickness in the MMP-2group was significantly lower than the TIMP-2and PBS group(P<0.05), and it was significantly difference between the PBS and TIMP-2group (P<0.05);④Masson trichrome collagen staining:10d,16d after surgery, the collagen fibers of the wound healing area was smaller and arranged in neat in the MMP-2group;in most cases collagen fibers was more disordered in the TIMP-2group; the thickness of collagen fibers in the PBS group was mixed and arranged in a little disorder(P<0.05).Conclusions:①In the wound healing period, the use of MMP-2can inhibit the differentiation of fibroblasts, inhibiting the inflammatory response, reducingcollagen synthesis, inhibiting of a-SMA expression, delayed wound healing; ultimately it can reduce scar formation after healing.②The use of MMP-2lead to delayed healing in local wound for Rats, but the scar formation will be reduced after healing. Reversely, the use of TIMP-2will promote healing and scar formation, so it can play the best role only to maintain the proper proportion of MMP-2and TIMP-2.
