Object:The goal of the study is to analyze the function and mechanism of PLC? in the wound healing.Futher,we investigate its role in wound healing rate as well as scar formation.18 PLC?gene knockout mice and wild type mice was divived in two independent groups.3mm cycle skin wound was made for all of them.Methods:1.Measure the size of wound for 6 mice of each group until healing completely to study the wound healing rate.2.Take the wound surround tissue and compare the cells by H&E staining.3.Stain the slide by PCNA and analyze the cell proliferation.4.Study the i-SMA?Col6?Col17 by immunostaining.5.Using the Lifeline Cell Technology as well as si RNA to knockout PLC?gene to study the function of PLC?gene in cell movement rate.Result:1.There is no clear difference for PLC?gene knockout mice and wild type mice in the wound healing rate.Meanwhile,the wound healing rate of Day 3,4,5PLC?gene knockout mice were inhibited.2.There is difference for two group in the epithelia cells rebuilding however,the cells of WT mice is more that PLC?gene knockout mice.3.There is difference for two group in the epithelia cells proliferation.4.The Col6 and Col17 of PLC?gene knockout mice is more than WT mice5.The cell migration rate of WT mice is larger than PLC?gene knockout mice.Conclusion:1.The PLC?has some function in inflammation and IL-6?Cxcl-1?Cxcl-2?Ccl20were the downstream factors for its.2.PLC? does not affect cell proliferation but has some function on its migration rate.3.By the two conclusions above,PLC?can delay the wound healing.4.PLC?can clearly redue the wound scar formation.
