"Zhilou Xunxi Ji" Formulation Optimization Of Experimental And Clinical Research

Objective:This research is aimed to optimize the "Zhilou Xunxi Ji" prescription. We designed several animal experiments to reform the medicine amount and dosage of Zhilou Xunxi Ji and so as to provide objective experimental data to demonstrate the basal treatment effect of the adjusted prescription of Zhilou Xunxi Ji. And according to the experimental data, we designed two trials with eliminating rhubarb or cuspidatum to demonstrate the clinical treatment effect. We attempt to discover feasibility of optimizing the prescription by the animal and clinical trials.Methods:1. Animal experiments:We designed the dropsy of big mouse anus verge trial, dropsical sole of foot trial, hot board and turning round body trials with small mouse, to compare the basal treatment effect, anti-inflammatory and relieving pain effect of prime Zhilou Xunxi Ji prescription with that of the groups of half dosage, eliminating rhubarb and eliminating cuspidatum.2. Clinical trials:40postoperative patients of mixed hemorrhoids were divided into two groups. The study group were treated with the Zhilou Xunxi Ji of eliminating cuspidatum. The controlled group use the prime prescription. Observing contrastively postoperative pain, dropsy and wound healing state.Results:1. Experimental results:the prime prescription, half dosage, eliminating rhubarb and eliminating cuspidatum all can effectively mitigate the dropsy of big mouse anus verge caused by croton oil-induced inflammation agent, dropsy of sole of foot caused by Carrageenan and reduce the content of PGE2. And they can also improve effectively mouse pain threshold and reduce the times of mouse turning round body caused by acetic acid. Pathologically, Zhilou Xunxi Ji can mitigate proctitis and anal canal inflammation. It is the best to mitigate dropsy and pain with the prime prescription. It is nonsignificant statistically to compare the effect of eliminating rhubarb with eliminating cuspidatum.2. Clinical results:Zhilou Xunxi Ji is better than eliminating cuspidatum in mitigate dropsy. Although other results is nonsignificant statistically, treatment effect of the prime prescription is better than that of eliminating cuspidatum overall from the data distribution.Conclusion:From the primary results, we can demonstrate that no matter to reduce dosage, eliminate cuspidatum or eliminate rhubarb, they are no the best way to optimize the Zhilou Xunxi Ji prescription. And we are expecting a further research to optimize the prescription.
