Objective: A number of studies have evaluated the correlationbetween interleukin-1A (IL-1A)+4845, interleukin-1B (IL-1B)-511gene polymorphism and aggressive periodontitis susceptibility correlation,but the results remain inconclusive. To get the future conclusion, weperformed this Meta-analysis.Methods: Systematic searches of Chinese Biomedical LiteratureDatabase (CBM), China Academic Journal database (CNKI), VIP,Wanfang Database, the Cochrane library,PUBMED, MEDLINE andEMBASE, ISI Web of Science database were performed to collect data oncase-control study and cohort study of IL-1A+4845interleukin,IL-1B-511gene polymorphism and aggressive periodontitis susceptibilitycorrelation.Meanwhile, we used search engines like Google to track thereferences and conducted a comprehensive retrieval. The retrieval timebegan from the database’s building to December2011. Experimentsincluded in the article are case-control experiments and cohort study,required similar research methods that must consist of the purpose, design and specific statistical methods. Its findings are available to gain odds ratio(odds ratio, OR) and95%CI (95%confidence interval,95%CI) or can beconverted into OR values and95%CI of the basic data. We evaluated thequality of the articles included in the study using the Newcastle-Ottawascale. Meta-analysis was carried out by software Review Manager (Version5.1.2). The combined effects used odds ratios (ORs) and95%confidenceintervals (95%CIs) to be evaluated. Publication bias was judged directly byfunnel plot. Sensitivity analysis was made to estimate the combined effectby repeating meta-analysis with each of the studies individually removed inorder.Results: Nine literature and ten studies were included in themeta-analysis ofIL-1A+4845gene polymorphism associated with the AP, which areincluding469persons in the case group and837persons in the controlgroup. In addition, nine literature and nine studies were included inmeta-analysis of IL-1B-511gene polymorphism associated with the AP,which are including581persons in the case group and740persons in thecontrol group. There are five literature includes both the IL-1A+4845genepolymorphisms and the relation between IL-1B-511gene polymorphismand the AP.1) the relationship between IL-1A+4845gene polymorphisms and APMeta-analysis results shows that: allele genetic model of genotype,T allele and G allele (OR=1.29,95%CI=1.03-1.61, P=0.03);the correlation between T allele of IL-1A+4845gene polymorphism andAP was statistically significant., the differences between additive geneticmodel of the genotype TT and GG (OR=1.43,95%CI=0.83-2.45, P=0.20) were not statistically significant; dominant model of genotype GT+TT and the GG (OR=1.32,95%CI=1.01-1.73, P=0.04); thedifferences between recessive genetic model of genotype TT and GT+GG(OR=2.16,95%CI=1.31-3.55,P=0.002)were statistically significant.In the race-based subgroup analysis,we analyzed the Caucasian andAsian race. Meta-analysis displayed significant association betweenIL-1A+4845gene polymorphism and susceptibility of Caucasian AP,while no evidence demonstrated that related to the Asian ethnic origin.2) the relationship between IL-1B-511gene polymorphisms and APMeta-analysis results shows that: allele genetic model ofgenotype,C allele and T allele (OR=1.22,95%CI=1.05-1.43, P=0.01);the correlation between C allele of IL-1B+511gene polymorphism and APwas statistically significant. The differences between additive geneticmodel of the genotype CC and TT (OR=1.05,95%CI=0.75-1.45, P=0.78) were not statistically significant; dominant model of genotype CT+CC and TT (OR=0.92,95%CI=0.71-1.20, P=0.53); the differencesbetween recessive genetic model of genotype CC and CT+TT (OR=1.09,95%CI=0.85-1.39,P=0.50)were not statistically significant. Meta-analysis displayed significant association between IL-1B-511gene polymorphism and susceptibility of Asian AP, while no evidencedemonstrated that related to the Caucasian race.Conclusions:1) Based on the results of the present study, threegenetic model’s Meta-analysis show that the IL-1A+4845genepolymorphism associates with the susceptibility of aggressiveperiodontitis. A genetic model displays the differences between TT andGG gene are not statistically significant. The description above illustratesthat IL-1A+4845gene polymorphism may be associated with aggressiveperiodontitis susceptibility.2) Allele genetic model of genotype show that the differencesbetween C allele and T allele are statistically significant. The other threegenetic models show that the differences of IL-1B-511gene polymorphismand aggressive periodontitis are not statistically significant, indicating thatIL-1B-511gene polymorphism may be associated with susceptibility toaggressive periodontitis.
