Objective:observe the effect that Danzikangxi granules influence the rat knee osteoarthritis chondrocytes apoptosis, and then investigate the efficacy that Danzikangxi Granules remedy knee osteoarthritis.Methods:choose80healthy Wistar rats with weight (250±10) g/one (male and female’s numbers are equal.); divide them into4groups in accordance with the random number table, each group has20rats. Group a is Danzikangxi granules group; group b is positive comparison group (western medicine group); group c is model group; group d is blank control group. Group a, b and c are caused rat knee osteoarthritis by used improvable Hulth modeling method. After6weeks, group a is poured Danzikangxi granules into their stomachs; group b is used subcutaneous injection with S-methyl isothiourea; group c and d are poured the equal volume of distilled water of traditional Chinese medicine. After4weeks, observe the general form of the rat knee joint in naked eyes, extract the fluid of the knee joint and detect NO content, use TUNEL detect the apoptosis rate of the joint chondrocytesResults:under eyes obeservation, model group’s modeling knee cartilage obviously degenerate, appears local ulcer and joint marginal hyperplasia; blank group’s knee cartilage is smooth and moist; Danzikangxi group and western medicine group’s modeling knee joints generate new cartilage. NO content of the Knee synovial fluid in normal group is lower than the other3groups (P<0.05); NO content of the Danzikangxi group and the western medicine group is lower than model group (P <0.05); there is no obvious difference between the Danzikangxi group and the western medicine group (P>0.05);the apoptosis rate of the Danzikangxi group and the western medicine group is obviously lower than model group (P <0.05), the apoptosis rate between the Danzikangxi group and the western medicine group has no obvious difference (P>0.05); the apoptosis rate of the Danzikangxi group, the western medicine group and the model group is higher than the blank group (P <0.05)Conclusion:Danzikangxi granules can reduce the NO content of the rat knee joint synovial fluid, and thus control the joint chondrocyte apoptosis, it has preventive and therapeutic effects for the early and mid-cartilage degeneration of the rat knee osteoarthritis.
