Objective: To research the expression and significance of Iroquoisgene1(IRX1) in human normal and carcinoma hepatic cell lines and livertissues. Help explore the early diagnosis and the prognosis monitoring ofspecific indicators of liver cancer and provide potential targets to find anddetermine the theoretical basis and experimental evidence for the targetedtherapy of liver tumors.Method: We examined the expression of Homeobox gene IRX1inhuman normal liver cell line LO2and hepatoma carcinoma cell linesSMMC-7721、Huh7、MHCC-97H、HepG2、 HepG2-215、QGY-7701, and20liver tissue samples which were stored in liquid nitrogen by fluorescentquantitative real-time polymerase chain reaction and western blot, and thenwe determined the localization and clinical characteristics of Homeoboxgene IRX1in93liver tissue samples which were embedded in paraffinblocks using immunohistochemistry.Result:(1) Compared with normal liver cell line LO2, the mRNA andprotein expressions of Homeobox gene IRX1were higher in the human hepatic cancer cell lines SMMC-7721、Huh7、MHCC-97H、HepG2、HepG2-215、QGY-7701. The amount differences between the differenttumor cell lines and normal hepatic cell lines were statistical significance (P<0.05).(2) Compared with non-carcinoma control liver tissues, the mRNAand protein expressions of Homeobox gene IRX1were higher in the humanprimary hepatocarcinoma tissues. The degree of tumor differentiation wasrelative with its expression level, the lower differentiation, the higher itsexpression. The differences between non-carcinoma control tissues andvarious differentiation hepatocarcinoma tissues were statistically significant(P <0.05); IRX1positive stain in the differentiation of human primaryhepatocellular carcinoma tissues by immunohistochemical staining resultswas significantly higher than non-cancerous control liver tissues,Furthermore, association analysis also revealed that the increasedHomeobox gene IRX1in tumor tissues is related to higher pathologicclassification (p﹤0.05).Conclusion: Those findings implicate that Homeobox gene IRX1probable regulate the process of tumorigenesis in human primaryhepatocellular carcinoma, which can be applicable to detect the presence ofhepatocarcinoma and be developed as an indicator of prognosis andtargeted therapy. IRX1is expected to become the new target of moleculartargeted therapy for primary liver cancer and improve the relevance andeffectiveness of the treatment of primary liver cancer and Clinical Oncology targeted therapy to indicate a new basis of the theoretical basisand a new direction for cancer treatment.
