Objective:To study the behavioral changes and expression ofinflammatory factors in chronic prostatitis rat models,and the differentexpression of5-HT and serotonin transporter in within T13-S1spinalsegments of chronic prostatitis rat models,and the effect of Escitalopram onchronic prostatitis rat model.Methods:Male Sprague Dawley rats were randomly divided into6groups, the control group, the45d model group, the60d model group, the90d model group, the treatment15d group and the treatment45d group.The chronic prostatitis rat model was made in the latter five groups byabdominal subcutaneous multiple point’s injection of the saline extract ofmale rat prostate glands(RPG) in Freund’s complete adjuvant(FCA) andPertussis-Diphtheria-Tetanus vaccine0.5ml i.p. at the0and30th day, andfrom the46th day of the experiment, the rats in the treatment15d groupand the treatment45d group were given intragastric injection of salinecontaining10mg.kg-1.d-1Escitalopram Oxalate Tablets respectively forcontinuous15days and45days. Two days before kill the rats, the behavioral change was determined by open-field test and sucroseconsumption test, the morphological change of prostatic tissue wasobserved by light microscopy, and the levels of interleukin10(IL-10),interleukin2(IL-2), interleukin1β (IL-1β) and tumor necrosisfactor-α(TNF-α) in blood serum and prostatic tissue were detected byenzyme linked immunosorbent assay(ELISA).The expression of5-HT andserotonin transporter in within T13-S1spinal segments was detected byimmunohistochemistry, The expression of SERT mRNA was detected byfluorescent quantitative real-time PCR.Results:The lymphocytes infiltrated prostates interstitium in45dchronic prostatitis rat models.Inflammatory reaction was intensity and agreat quantity of lymphocytes were observed around the vessels in60dchronic prostatitis rat models.Segmental necrosis were observed in Prostateepithelium of90d model group. Compared with the normal control group,the crossing movement、rearing movement times of open-field test andsucrose consumption were significantly decreased in the90d model groupand no significant difference in the45d model group、60d model group、the treatment15d group and the treatment45d group.Compared with thenormal control group, the serum and prostatic expression levels ofinflammation factors such as IL-10、IL-1βand TNF-α in the60d and90dmodel group were increased considerably.Compared with the60d modelgroup,the prostatic and the serum levels of IL-1βwere significantly decreased and IL-10were significantly higher in the treatment15d group;Compared with the90d model group,the prostatic levels of IL-10wassignificantly higher and the serum levels of IL-1βwas significantly reducedin the treatment45d group.Compared with the normal control group,theaverage gray scale of5-HT increased markedly in L3-L4spinal segmentsof the45d model group、dorsal horn of T13-L2spinal segments in the60dmodel group、T13-L2spinal segments、dorsal horn of L3-L4spinalsegments and anterior horn of L5-S1spinal segments in the90d modelgroup.Compared with the normal control group,the average gray scale ofSERT in L5-S1spinal segments of the90d model group increasedmarkedly.Compared with the normal control group,the average gray scaleof5-HT in L3-L4spinal segments of the treatment15d group increasedmarkedly.Compared with the normal control group,the average gray scaleof SERT in the anterior horn and dorsal horn of T13-L4spinal segmentsdecreased significantly of the treatment45d group. Compared with the90dmodel group,the average gray scale of SERT in the anterior horn anddorsal horn of T13-S1spinal segments and the average gray scale of5-HTin the dorsal horn of T13-L2spinal segments and the anterior horn ofL5-S1spinal segments decreased significantly.Compared with the normalcontrol group,the SERT mRNA level of T13-L2spinal segments decreasedsignificantly in the45d、60d、90d model group and the treatment15d、45d group;the SERT mRNA level of L3-L4spinal segments decreased significantly in the45d、60d model group and the treatment15d group;the SERT mRNA level of L5-S1spinal segments decreased significantly inthe60d、90d model group,the SERT mRNA level of L5-S1spinalsegments increased significantly in the treatment45d group. Comparedwith the60d model group,the SERT mRNA level of L5-S1spinalsegments increased significantly in the treatment15d group.Comparedwith the90d model group,the SERT mRNA level of T13-L2spinalsegments decreased significantly and the SERT mRNA level of L5-S1spinal segments increased significantly.all with statistical significance.Conclusions:chronic prostatitis model rats exhibited characteristics ofdepression.The level of inflammatory factors TNF-α、IL-1βand lL-10wereup-regulation in chronic prostatitis model rats.Our data indicated a distinctexpression of the average gray scale of5-HT and SERT and the SERTmRNA level of SERT in the T13-S1spinal segments at differentstages,suggesting5-HT and SERT may be play a role of regulation in theprosess of chronic prostatitis.The treatment of Escitalopram OxalateTablets could reduce the prostatic inflammatory response of model rats,may play the function of improving behavior,resisting depression in rats,and regulate the local immune condition and the expression level of5-HT、SERT and SERT mRNA in the T13-S1spinal segments....
