Objective:In order to explore chronic prostatitis basic pathogenesis,we study the pathological mechanisms from cell signal level,preliminary clarified the treatment and mechanism of qian lie kang tai formula for chronic bacterial and non-bacterial prostatitis,preliminary revealed the scientific connotation about the theory of "liver really and kidney weak",Method:1.Observed the inhibition of qian lie kang tai formula to mice ear swelling caused by croton oil and rat foot swelling caused by jiao cha cai colloidal.2.Observed the effection of qian lie kang tai formula to rat prostate tissue morphology changement in chronic bacterial and non-bacterial prostatitis.The experiment was divided into two parts.①Established the model of chronic bacterial prostatitis:Narcoticed 45 wistar rats,we opened abdominal skin and muscle under aseptic conditions,injected the concentration of 107~109/ml E.coli suspension 0.2ml into two side of rat prostates,finally sutured abdominal muscle and skin.The 45 rats were randomly divided into chronic bacterial prostatitis model group,levofloxacin group and qian lie kang tai formula group,each group of 15 rats.②Established the model of chronic non-bacterial prostatitis:Narcoticed 45 wistar rats,we opened abdominal skin and muscle under aseptic conditions,injected xiao zhi ling injection 0.2ml into two side of rat prostate,finally sutured abdominal muscle and skin.The 45 rats were randomly divided into chronic non-bacterial prostatitis model group,qian lie kang group and qian lie kang tai formula group,each group of 15 rats.Finally narcoticed 15 wistar rats,we opened abdominal skin and muscle under sterile conditions,injected aseptic N.S 0.2ml into two side of rat prostate,sutured abdominal muscle and skin.This 15 rats is the common control group for two experiments.Each group was given corresponding drugs after the operation 7 days,2 times a day,for 5 weeks.Observe the morphology changement of each group prostate tissue. 3.Under the electron microscope,observe the effection of qian lie kang tai formula to chronic bacterial and non-bacterial prostatitis rat prostate cell structure.4.Used radiation immunity technology to detect the lever of each group's rat plasma TXB2 and 6-keto-PGF1α.5.Used enzyme-linked immunnosorbnent assay technology to detect the level of each group's rat prostate tissue TNF-αand IL-1β.6.Used reverse transcription-polymerase chain reaction technology to detect the level of each group's rat prostate tissue TNF-αand IL-1βmRNA.Resulut:1.Qian lie Kang tai formula had the obvious inhibition for mice ear swelling and rat foot swelling(P<0.05).2.The changement of rat prostate tissue under morphology:In chronic bacterial and non-bacterial prostatitis model groups,the secretion of prostate epithelial cells were reduced,inflammatory cells and collagen fibers hyperplasia between epithelial cells were increased.Pathological damagement was obviously.Inflammatory cells and collagen fibers hyperplasia between epithelial cells were reduced in qian lie kang tai formula group.3.The changement of rat prostate cell structure under electron microscopy:In two model groups,there were obvious endoplasmic reticulum vesiculation,mitochondrial swelling and collagen fibers hyperplasia in prostate epithelial cells.The endoplasmic reticulum vesiculation,mitochondrial swelling and collagen fibers hyperplasia significantly were reduced in qian lie kang tai formula group.4.Observed the lever of each group rat plasma TXB2 and 6-keto-PGF1αin chronic bacterial and non-bacterial prostatitis:Compared to the normal group,the levels of rat plasma TXB2 were increased,6-keto-PGF1αwere reduced in two model groups(P<0.05),the ratioes of TXB2/6-Keto-PGF1αwere significantly increased(P<0.01). Compared to the model group,the levels of rat plasma TXB2 were reduced, 6-keto-PGF1αwere increased in qian lie kang tai formula group(P<0.05),the ratioes of TXB2/6-Keto-PGF1αwere significantly reduced(P<0.01).5.The changement of rat prostate tissue cytokines TNF-αand IL-1βin chronic bacterial and non-bacterial prostatitis:Compared to the normal group,the levels of rat prostate tissue cytokines TNF-αand IL-1βin two model groups were increased(P<0.05).Compared to the model group,the levels of rat prostate tissue cytokines TNF-αand IL-1βin qian lie kang tai formula group were reduced(P<0.05).6.The changement of rat prostate tissue cytokine TNF-αand IL-1βmRNA in chronic bacterial and non-bacterial prostatitis:Compared to the normal group,the levels of rat prostate tissue cytokines TNF-αand IL-1βmRNA in two model groups were increased(P<0.05).Compared to the model group,the levels of rat prostate tissue cytokine TNF-αand IL-1βmRNA in qian lie kang tai formula group were reduced(P<0.05).Conclusion:The study raised that the theory of "liver really and kidney weak" was chronic prostatitis pathology features.Through this experiment we found that qian lie kang tai formula could reduce animal's inflammation,had the obvious anti-inflammation function,significantly reduced the pathology injury under microscopy.Compared to the normal group,the levels of rat plasma TXB2 in chronic bacterial and non-bacterial prostatitis model groups were increased,6-keto-PGF1αwere reduced,the ratioes of TXB2/6-Keto-PGF1αwere increased.It showed that blood stasis existed in chronic prostatitis.Compared to the model group,qian lie kang tai formula could reduce the levels of rat plasma TXB2,increase the levels of rat plasma 6-keto-PGF1α, significantly reduce the ratioes of TXB2/6-Keto-PGF1α.It showed that qian lie kang tai formula could improve blood stasis,which may be the one of mechanism for chronic prostatitis.The levels of rat prostate tissue cytokines TNF-α,IL-1αand TNF-α,IL-1βmRNA were increased in chronic bacterial and non-bacterial prostatitis model groups,which showed that there were indeed abnormal immune response in chronic prostatitis,this abnormal immune response may be common pathogenesis of chronic bacterial and non-bacterial prostatitis.Qian lie kang tai formula could reduce the levels of rat prostate tissue cytokines TNF-α,IL-1βand TNF-α,IL-1βmRNA. It showed that qian lie kang tai formula had a certain adjustment role to prostate tissue cytokines and their receptors,which may be the one of mechanism for chronic prostatitis.
