The Clinical Significance And Expression Of Paf And TNF-α And PAF-rin Semen Of Leukocyto Spermic Patients

ObjectiveTo observe the expression and correlation of platelet activation factor(PAF) and its receptor (PAF-R) and tumor necrosis factor-α(TNF-α) inLeukocytospermia， to discuss the role and mechanism about plateletactivate factor (PAF) and its receptor (PAF-R) in the genital tractinflammation.MethodsCollect our hospital urology clinic of male patients, the semen routineexamination and peroxidase dyeing method was used to determine thesperm.22cases of leukocytospermia patients for the case group,15casesof normal men as control group. The platelet active factor (PAF) and tumornecrosis factor-α(TNF-α) level in the seminal plasma was detected byELISA, the expression of platelet active factor receptor (PAF-R) in spermcells and the white blood cells was detected by immune cells chemicaltesting.Results The PAF of leukocytospermia were (2.14士0.43) ng/ml, which weresignificantly lower than the normal group (6.21士1.38) ng/ml (P <0.01);the TNF-α(5.51士1.46) ng/ml significantly higher than normal semengroup (3.48士1.08) ng/ml, the level of TNF-α was negative correlation toPAF (r=-0.68, P <0.01), the level of PAF was negative correlated to theWBC (r=-0.62, P <0.01), the TNF-α was positive associated with WBC(r=0.77, P <0.01). By immune cell chemistry, the expression of thePAF-R in leukocytospermia and normal sperm were(1.32士0.36) and(2.68士0.45), the expression of PAF-R in white blood cells about two groups,respectively (1.08士0.18) and (0.36士0.12),(p <0.01).Conclusion1.The lower expression of platelet active factor (PAF) and the highexpression of tumor necrosis factorα (TNF-α) in leukocytospermia diseasemay affect the semen quality.2. The PAF was increased in general inflammation,the lower expressionof PAF when leukocytospermia shows that its particularity and importantscientific significance, the reason is not clear.3.To compared with normal the reduction of PAF-R in sperm cell ofleukocytospermia and the increase of PAF-R in white blood cells ofleukocytospermia may make semen quality drop，the specific mechanismneed to be studied.
