Objective:to investigate the expression level of EZH and P27in benign prostatic hyperplasia(BPH) and prostate cancer(PCa) and also analysis their correlation with clinical staging and pathological grading, in addition to assess their diagnostic efficacy and prognostic value.Methods:this retrospective study comprises90surgically resected prostate tumor specimens between January2007and December2011, among them58cases were PCa and34cases were BPH. After sectioning of formalin fixed paraffin-embedded specimens, the two steps IHC method were used for immunohistochemical analysis. Finally all slides were reviewed by tow specialist. Fisher’s exact test and Pearson’s correlation were used to analysis the relationship among clinical staging, pathological grading and antibody expression.Results:1-EZH expression rate were41.2%(7/17),86.7%(26/30) in BPH and PCa respectively and the difference was statistically significant (P<0.05).2-P27expression rate were76.5%(13/17),43%(12/28) in BPH and PCa respectively, the difference between them was statistically significant (P<0.05).3-In this study, the expression rate of EZH and P27had statistically significant correlation with clinical staging and pathological grading.Conclusion:1-EZH expression level was significantly higher in PCa then BPH also it had a positive correlation with clinical staging and pathological grading; it means that the EZH will be a new biomarker for diagnosis of PCa.2-P27expression level was significantly high in BPH and also its expression level has an inverse correlation with clinical staging and pathological grading, it indicates that P27will improve the diagnosis of PCa.
